摘要
目的探讨宫瘤消胶囊对子宫肌瘤模型大鼠雌、孕激素水平及雌激素受体(ER)、孕激素受体(PR)表达的影响。方法将SD大鼠随机分为正常对照组、模型组、宫瘤消小、中、大剂量组,各组10只。宫瘤消小、中、大剂量组各给予宫瘤消胶囊0.27 g/kg、0.54 g/kg和1.08 g/kg,除正常对照组外,其余各组采用雌、孕激素负荷法建立大鼠子宫肌瘤模型,分别连续灌胃4周。观察各组大鼠血清雌、孕激素水平及子宫肌层ER、PR的表达。结果宫瘤消小、中、大剂量组雌二醇水平分别为(321.49±23.40)pmol/L、(277.19±12.00)pmol/L、(259.90±12.60)pmol/L;孕酮水平分别为(87.51±16.05)nmo/L、(72.94±7.11)nmo/L、(66.04±9.11)nmo/L;ER表达率分别为(50.60±10.23)%、(23.61±7.46)%和(21.61±6.46)%,PR表达率分别为(59.28±7.37)%、(33.61±6.02)%和(30.61±6.46)%。其中宫瘤消小剂量组各指标与模型组比较,差异均无统计学意义(P>0.05),而宫瘤消大剂量组各指标与模型组比较,差异均有统计学意义(P<0.05)。结论宫瘤消胶囊可以降低雌二醇、孕酮水平,影响靶器官中ER、PR的表达,可能是宫瘤消胶囊抑瘤、缩瘤的机制之一。
Objective To observe the effect of Gongliuxiao capsule on the level of estrogen and progesterone and expression of theirs receptors (ER and PR) in myometrium of uterine leiomyomas modle rats. Methods SD rats were randomly divided into 5 groups, include control group, model group, low-dosage Gongliuxiao capsule group, middle-dosage Gongliuxiao capsule group and high-dosage group. Except for normal control group, the rats in the others groups were given estrogen and progesterone load to induce uterine leiomyomas. After continuous oral administration for 4 weeks, the concentration of estrogen and progesterone and the expressions of ER and PR in myometrium of uterine were detected. Results After treatment, compared with model group, there was obviously reduce of both the concentration of estrogen and progesterone and expressions of ER and PR in the low, middle and high dosage group. The concentration of estrogen separately was (321.49 ±23.40) pmol/L. (277.19 ±12.00) pmol/L. (259.90±12.60) pmol/L; progesterone (87.51± 16.05) nmo/L. (72.94±7.11) nmo/L. (66.04±9.11) nmo/L. The expression of ER was (50.60± 10.23) %, (23.61 ±7.46) % and (21.61 ±6.46) %.The expression of PR was (59.28±7.37) %, (33.61 ±6.02) % 和 (30.61 ±6.46) %, respectively.It was showed that there were no statistical differences between low-dosage group and model group, but had a decline trend (P 〉 0.05). The most significant changes of the concentration of estrogen and progesterone and expressions of theirs receptors were detected in high-dosage group (P 〈 0.05). The high-dosage Gongliuxiao capsule make the concentration of estrogen decline 95.68 pmol/L, progesterone decline to 80.93 nmo/L and the expressions of ER decline to 37.46%, PR drop to 35.67%. Conclusions The possible mechanism of Gongliuxiao capsule inhibition and reduce of uterine leiomyomas is adjusting the concentration of estrogen and progesterone, impacting the content of ER, PR in target organs.
