摘要
目的探讨黄芪葛根汤有效组分及其配伍对糖尿病大鼠肝脏脂代谢及炎症反应的作用机理。方法将86只SD大鼠随机分为正常组、模型组、金芪降糖片组、总黄酮组、总皂苷组、总黄酮与总皂苷组。一次性腹腔注射STZ(按46 mg·kg^(-1))诱导糖尿病大鼠模型。生化分析仪检测TG、CHO的水平,Real-time PCR法检测肝组织AdipoR1、AMPK、P38MAPK、PPARγ、LXRα、SREBP1mRNA表达。结果同正常组比较,模型组各项指标均有统计学意义(P<0.05)。与模型组比较,总黄酮能显著降低血清中CHO的浓度;总黄酮、总皂苷均能降低TG水平,且能增强AdipoR1、LXRαmRNA降低P38MAPK、SREBP1mRNA的基因表达。对于促进AMPK、PPARγmRNA基因的表达,总黄酮的作用更显著,总皂苷及两药的交互作用不明显。结论总黄酮发挥降脂抗炎作用可能主要与AMPK、PPARγmRNA表达有关;总皂苷的降脂抗炎作用可能主要与P38MAPKmRNA表达有关;两组分配伍的作用可能与激活AMPK/SREBP1信号传导通路有关。
Objective To explore the mechanism of the effective components and compatibility from Huangqi Gegen Decoction(黄芪葛根汤)on liver lipid metabolism and inflammatory response in diabetic rats.Methods Eight-six SD rats were randomly divided into normal group,model group,Jinqi Jiangtang Tablet group,total flavonoids group,total saponins group,total flaonoids and total saponins group.Diabetic rat model was induced by intraperitoneal injection of STZ(46 mg·kg^(-1)).The concentrations of TG and CHO in serum were detected by using biochemistry analyzer.Real-time PCR was used to detect the expressions of AdipoR1,AMPK,P38 MAPK,PPARγ,LXRαand SREBP1 mRNA in liver tissue.Results Compared with those of the normal group,the indicators in the model group were statistically different(P<0.05).Compared with the model group,total flavonoids can significantly reduce the concentration of CHO in serum.Total flavonoids and total saponins can decrease the levels of TG in serum,and can enhance the expressions of AdipoR1 and LXRαmRNA,but weaken the expressions of P38 MAPK and SREBP1 mRNA.For the promotion of expressions of AMPK and PPARγmRNA,the effect of total flavonoids was more significant.Total saponins and the interaction between total flavonoids and total saponinsis was not obvious.Conclusion The lipid-lowering and anti-inflammatory effects of total flavonoids may be related to the expressions of AMPK and PPARγmRNA.The lipid-lowering and anti-inflammatory effects of total saponins may be related to the expression of P38 MAPK mRNA.The role of total flavonoids in combination with total saponins may be related to the activation of AMPK/SREBP1 signaling pathway.
