摘要
为提高杂交兰种苗繁殖效率,提升种苗品质,以春兰与大花蕙兰杂交后代为试验材料,通过对其类原球茎(protocorm-like body,PLB)增殖与分化、生根壮苗等培养的研究。结果表明:(1)在1/2MS基本培养基中添加6-BA 0.5 mg/L+NAA 1.0 mg/L有利于杂交兰类原球茎增殖与分化。(2)三种防褐化物质中活性炭为抑制类原球茎褐化的主要影响因子。(3)在花宝二号基本培养基中添加IBA 0.5 mg/L+6-BA 1.0 mg/L有利于杂交兰组培苗生长。(4)不同有机添加物中椰汁为杂交兰组培苗生长的主要影响因子。综上,本研究为建立杂交兰K6组培快繁体系及其优良种苗规模化生产提供科学依据。
In order to speed up the propagation efficiency of Cymbidium hybrid seedlings and improve the quality,the hybrid progeny of Cymbidium hybridum and Cymbidium goeringii were used as experimental materials,through the study on the proliferation and differentiation,rooting and tissue seedling culture of PLB.The results showed:(1)Adding 6-BA 0.5 mg/L+NAA 1.0 mg/L to 1/2 MS basic medium was beneficial to the proliferation and differentiation of Cymbidium hybrid PLB.(2)Among the three anti browning substances,active carbon was the main factor to inhibit the browning of PLB.(3)IBA 0.5 mg/L+6-BA 1.0 mg/L was beneficial to the growth of Cymbidium hybrid tissue culture seedlings.(4)Coconut juice has great influence on the growth of Cymbidium hybrid tissue culture seedlings.Above all,the study provided scientific evidence for the large scale production and the establishment of tissue culture and rapid propagation system of Cymbidium hybrid.
