摘要 目的:探讨MicroRNA在动脉粥样硬化临床诊断中的作用。方法:选择本院120名冠状动脉粥样硬化的患者(冠脉粥样硬化组),另选择45名健康者为健康对照组,检测两组血脂,血清白细胞介素(IL)-6、IL-8、肿瘤坏死因子(TNF)-α水平以及microRNA-155、micro RNA-143、microRNA-145的表达,并分析冠脉粥样硬化患者上述microRNA表达与血脂、炎症因子水平间的相关性。结果:与健康对照组比较,冠脉粥样硬化组血清microRNA-155表达显著升高,microRNA-143,microRNA-145表达显著降低(P均=0.001);且冠状动脉粥样硬化患者中microRNA-155表达与IL-6、IL-8、TNF-α、TC、LDL-C水平均呈显著正相关(r=0.627~0.737,P均=0.001),与HDL-C水平呈显著负相关(r=-0.668,P=0.001);microRNA-143(r=-0.593~-0.458)和microRNA-145(r=-0.783~-0.311)表达与IL-6、IL-8、TNF-α、TC、LDL-C水平呈显著负相关(P均=0.001),与HDL-C水平均呈显著正相关(r=0.572、0.824,P均=0.001)。结论:脂质代谢及血管炎症反应参与了动脉粥样硬化的进展,microRNA-155、microRNA-143、microRNA-145与之密切相关,且对动脉粥样硬化的临床诊断具有重要的意义。 Objective:To explore role of microRNA in clinical diagnosis of atherosclerosis(AS).Methods:A total of 120 patients with coronary AS(CAS)from our hospital were selected as CAS group,another 45 healthy subjects were selected as healthy control group.Levels of blood lipids,serum interleukin(IL)-6,IL-8 and tumor necrosis factor(TNF)-α,and expressions of microRNA-155,microRNA-143 and microRNA-145 were measured and compared between two groups,and correlation among levels of blood lipids,inflammatory factors and above microRNA expressions were analyzed in CAS patients.Results:Compared with healthy control group,there was significant rise in serum microRNA-155 expression,and significant reductions in expressions of microRNA-143 and microRNA-145 in CAS group,P=0.001 all;in CAS patients,microRNA-155 expression was significant positively correlated with levels of IL-6,IL-8,TNF-α,TC and LDL-C(r=0.627~0.737,P=0.001 all),and significant inversely correlated with HDL-C level(r=-0.668,P=0.001);expressions of microRNA-143(r=-0.593~-0.458)and microRNA-145(r=-0.783~-0.311)were significant inversely correlated with levels of IL-6,IL-8,TNF-α,TC and LDL-C(P=0.001 all),and significant positively correlated with HDL-C level(r=0.572,0.824,P=0.001 both).Conclusion:Lipid metabolism and vascular inflammation are involved in AS progress.MicroRNA-155,microRNA-143,microRNA-145 are closely correlated with it,and they possess important significance for clinical diagnosis of AS.
