摘要
目的:分析溃疡性结肠炎(UC)的免疫细胞浸润情况。方法:从GEO数据库下载GSE11223微阵列测序数据集,样本通过Agilent-012391全人类基因组Oligo微阵列测序,利用R软件和免疫细胞浸润估计分析工具——反卷积算法(CIBERSORT),全面分析UC免疫浸润细胞。22个免疫细胞亚群的比例被评估,免疫浸润在配对正常组织和UC组织之间存在显著差异,这种差异可以表现为特征个体差异。在研究的细胞亚群中,使用免疫细胞比例进行的无监督聚类分析显示了5个亚群。结果:与正常组织巨噬细胞M2(P=0.014)、静息树突状细胞(P=0.012)高表达相比,UC参与固有免疫的特征性细胞有单核细胞(P=0.037)、巨噬细胞M0(P=0.001)、中性粒细胞(P=0.011),UC参与特异性免疫特征性细胞有CD4+记忆性T细胞(P=0.003)、调节性T细胞(P=0.034)。细胞亚群1、细胞亚群2的活跃,细胞亚群3和细胞亚群4功能的失调,导致了UC的发生发展。结论:CD4+记忆性T细胞、巨噬细胞M2型、调节性T细胞和UC的发生发展密切相关,而这些差异可能是药物治疗作用效应的重要决定因素。
Objective:To analyze the infiltration of immune cells in ulcerative Colitis(UC).Methods:The GSE11223 microarray sequencing data set was downloaded from the GEO database.The samples were sequenced by Agilent-012391 Oligo microarray sequencing of the entire human genome.The immune infiltrating cells of ulcerative colitis were comprehensively analyzed using R software and the immune cell infiltration estimation and analysis tool-deconvolution algorithm(CIBERSORT).The proportion of 22 immune cell subsets was assessed,and there was a significant difference in immune infiltration between paired normal tissue and ulcerative colitis tissue,which could be characterized by individual differences.In the cell subsets studied,unsupervised cluster analysis using the proportion of immune cells revealed five subsets.Results:Compared with the high expression of macrophage M2(P=0.014)and resting dendritic cells(P=0.012)in normal tissues,the characteristic cells of ulcerative colitis that participate in innate immunity were monocytes(P=0.037),macrophage M0(P=0.001)and neutrophils(P=0.011).Ulcerative colitis was characterized by the involvement of CD4+memory T cells(P=0.003)and regulatory T cells(P=0.034).The activity of cell subsets 1 and 2,and the dysfunction of cell subsets 3 and 4 lead to the development of ulcerative colitis.Conclusion:our results show that memory CD4+T cells,macrophages M2,and regulatory T cells are closely related to the development of ulcerative colitis,which may be important determinants of effect of drug treatment.
