基于生物信息学的子宫内膜癌诊断标志物的筛选及免疫细胞浸润分析 被引量:4

Screening of diagnostic markers for endometrial cancer and analysis of immune cell infiltration based on bioinformatics

摘要 目的:筛选子宫内膜癌(EC)的诊断生物标志物,并分析EC中免疫细胞浸润情况。方法:从GEO和TCGA数据库下载EC的表达谱数据集,利用R软件鉴定EC与正常子宫内膜组织的差异表达基因(DEGs),对其进行功能和通路富集分析。构建蛋白质-蛋白质相互作用(PPI)网络,筛选关键(hub)基因,并对hub基因进行ROC分析筛选出EC的诊断生物标志物。利用科学的反卷积算法(CIBERSORT)分析EC中免疫细胞浸润情况。结果:共鉴定出448个DEGs和10个hub基因。DEGs主要涉及染色体分离、核分裂等生物学过程,介导趋化因子活性、丝氨酸肽酶活性等分子功能,富集于着丝粒、细胞间桥和细胞外基质。富集的通路主要与“Wnt信号通路”“TGF-β信号通路”“细胞周期信号通路”及“HIF-1信号通路”相关。hub基因ROC分析结果显示AURKA、NCAPG、CDC20、CENPF和UBE2C具有较高的诊断价值。免疫细胞浸润分析发现CD4+记忆性T细胞和CD8+T细胞可能参与EC发生发展。结论:AURKA、NCAPG、CDC20、CENPF和UBE2C可作为EC的诊断标志物,CD4+记忆性T细胞和CD8+T细胞与EC发生发展密切相关,为EC的分子机制研究提供了新思路。 Objective:To screen diagnostic biomarkers of endometrial carcinoma(EC)and to analyze immune cell infiltration in EC.Methods:Expression profiles of EC were downloaded from GEO and TCGA databases,and differentially expressed genes(DEGs)of EC and normal endometrial tissues were identified by R software,and functional and pathway enrichment analysis were performed.Construct a protein-protein interaction(PPI)network,screen hub gene,and perform ROC analysis on hub gene to screen out diagnostic biomarkers of EC.Scientific deconvolution algorithm(CIBERSORT)was used to analyze immune cell infiltration in EC.Results:A total of 448 DEGs and 10 hub genes were identified.DEGs mainly involve biological processes such as chromosome segregation and nuclear division,and mediate molecular functions such as chemokine activity and serine peptidase activity,and are enriched in centromeres,intercellular bridges and extracellular matrices.Enriched pathways are primarily involved in"Wnt signaling pathway""TGF-βsignaling pathway""cell cycle signaling pathway"and"HIF-1 signaling pathway".ROC analysis of hub gene showed that AURKA,NCAPG,CDC20,CENPF and UBE2C have high diagnostic value.Immune cell infiltration analysis revealed that CD4+memory T cells and CD8+T cells may be involved in occurrence and development of EC.Conclusion:AURKA,NCAPG,CDC20,CENPF and UBE2C can be used as diagnostic markers for EC.CD4+memory T cells and CD8+T cells are closely related to development of EC which provides new ideas for molecular mechanism study of EC.
