摘要
目的探究人参皂苷Rg3对宫颈癌HeLa细胞增殖抑制和凋亡的影响机制。方法体外培养宫颈癌HeLa细胞;采用MTT法检测人参皂苷Rg3对宫颈癌HeLa细胞的抑制效果;将HeLa细胞分为对照组、低剂量Rg3组、中剂量Rg3组和高剂量Rg3组,分别采用0、50、100、200μg/ml的人参皂苷Rg3处理;采用克隆形成实验检测各组HeLa细胞增殖情况;采用流式细胞仪检测各组细胞凋亡情况;采用蛋白质印迹法检测各组增殖相关蛋白Ki67、PCNA、凋亡蛋白Bcl-2和Bax以及PI3K/Akt通路相关蛋白的表达情况。结果 MTT结果显示,随着人参皂苷Rg3浓度的增加,宫颈癌HeLa细胞增殖抑制显著升高(P<0.05);与对照组比较,低剂量Rg3组PCNA蛋白、Ki67蛋白、细胞克隆形成率、Bcl-2蛋白、PI3K蛋白和Akt蛋白相对表达水平均显著降低,Bax蛋白相对表达水平显著升高(均P<0.05);与低剂量Rg3组比较,中剂量Rg3组PCNA蛋白、Ki67蛋白、细胞克隆形成率、Bcl-2蛋白、PI3K蛋白和Akt蛋白相对表达水平均显著降低,Bax蛋白相对表达水平显著升高(均P<0.05);与中剂量Rg3组比较,高剂量Rg3组PCNA蛋白、Ki67蛋白、细胞克隆形成率、Bcl-2蛋白、PI3K蛋白和Akt蛋白相对表达水平均显著降低,Bax蛋白相对表达水平显著升高(均P<0.05)。结论人参皂苷Rg3可以抑制宫颈癌HeLa细胞增殖并促进其凋亡,作用机制可能为抑制PI3K/Akt信号通路,调控其下游相关增殖和凋亡蛋白的表达,且在一定范围内呈浓度依赖性。
Objective To explore the effect mechanism of ginsenoside on proliferation inhibition and apoptosis of cervical cancer(CC) HeLa cells.Methods CC HeLa cells were cultured in vitro.The inhibitory effects of ginsenoside Rg3 on CC HeLa cells were detected by MTT.HeLa cells were divided into control group,low-dose,medium-dose and high-dose Rg3 groups,respectively.The low-dose,medium-dose and high-dose Rg3 groups were respectively treated with 0,50,100 and 200 μg/ml ginsenoside Rg3.The proliferation of HeLa cells in each group was detected by clone formation assay.The apoptosis in each group was detected by flow cytometry.The expression of proliferation-related proteins Ki67,PCNA,apoptosis proteins Bcl-2,Bax and PI3 K/Akt pathway-related proteins in each group was detected by Western blot.Results MTT results showed that with the increase of ginsenoside Rg3 concentration,inhibition effects on CC HeLa cells proliferation were significantly increased(P<0.05).Compared with control group,PCNA,Ki67,clone formation rate of cells,relative expression levels of Bcl-2,PI3 K and Akt proteins were significantly decreased(P<0.05),while relative expression level of Bax protein was significantly increased in low-dose Rg3 group(P<0.05).Compared with low-dose Rg3 group,PCNA,Ki67,clone formation rate of cells,relative expression levels of Bcl-2,PI3 K and Akt proteins were significantly decreased(P<0.05),while relative expression level of Bax protein was significantly increased in medium-dose Rg3 group(P<0.05).Compared with medium-dose Rg3 group,PCNA,Ki67,clone formation rate of cells,relative expression levels of Bcl-2,PI3 K and Akt proteins were significantly decreased(P<0.05),while relative expression level of Bax protein was significantly increased in high-dose Rg3 group(P<0.05).Conclusion Ginsenoside Rg3 can inhibit the proliferation of CCHeLa cells and promote their apoptosis.The mechanism of action may be inhibiting PI3 K/Akt signaling pathway,regulating the expression of downstream related proliferation and apoptosis proteins,and
