摘要
目的研究HER-2/neu(c-erbB-2)和Ras癌基因在宫颈腺癌和鳞癌中的表达及意义,探讨其在宫颈癌发生发展中的作用.方法采用免疫组化SP法检测正常宫颈组织20例、宫颈腺癌40例及鳞癌45例中P185c-erbB-2和P21ras蛋白的表达.结果在正常宫颈组织中P185c-erbB-2和P21ras蛋白均不表达,在鳞癌中阳性表达率分别为13.3%和28.9%,在腺癌中阳性表达率分别为27.5%和55.0%.P21ras在宫颈腺癌及鳞癌组表达均显著高于正常组(P<0.0125),同时腺癌组明显高于鳞癌组;P185c-erbB-2在腺癌中的阳性表达率较正常组高(P<0.0125),鳞癌组与正常组及腺癌组与鳞癌组相比差异不显著(P>0.0125).P185c-erbB-2和P21ras蛋白的阳性表达率与细胞分化级别有明显相关性(P<0.0125),与临床分期、淋巴结转移无关;两者在宫颈癌组织中的表达显著相关(P<0.01,kappa=0.405,χ2=53.21).结论在宫颈癌中P185c-erbB-2和P21ras蛋白同时过表达可能是宫颈癌的发病机制之一,过表达现象在宫颈腺癌中尤为突出.
AIM: To detect the expressions of HER-2/neu (c- erbB-2) and Ras oncogenes in cervical carcinoma, and to evaluate the roles of HER-2/neu and Ras in the genesis and development of cervix squamous cell carcinoma and adenocarcinoma. METHODS: The SP immunohistochemical method was used to detect the expressions of P185^c-erbB-2 and P21^ras proteins in 20 cases of normal cervix tissue, 40 cases of adenocarcinoma and 45 cases of squamous cell carcinoma. RESULTS: P185^c-erbB-2 and P21^ras proteins were not observed in normal cervixes. Positive expression rates of the two proteins were 13.3% , 28.9% in cervix squamous cell carcinoma, and 27.5% , 55.0% in cervix adenocarcinoma. Compared with normal cervix tissue, P21^ras positive expression in adenocarcinoma and squamous cell carcinoma was different significantly ( P 〈0. 0125 ) , and the expression between adenocarcinoma and squamous cell carcinoma had obvious difference ;The positive expression of P185^c-ebB-2 in adenocarcinoma was higher than the normal tissue significantly (P 〈 0. 0125 ). The difference between squamous cell carcinoma and normal tissue, or adenocarcinoma and squamous cell carcinoma was not significant (P 〉 0. 0125). P185^c-erbB-2 and P21^ras proteins were correlated with the differentiation degree of carcinoma closely (P 〈 0.0125). No correlation was found between positive expression and clinical stage, nor lymphatic metastasis. In addition, there was an obvious correlation between P185^c-erbB-2 and P21^ras in cervix cancer(P 〈0.01,kappa =0.405,)X^2 =53. 21). CONCLUSION: P185^c-erbB-2 and P21^ras proteins were overexpressed in cervix cancer at the same time, especially in cervical adenocarcinoma, which may be one of mechanisms of carcinogenesis.
