摘要
目的:探讨黄连素对高糖诱导的内皮祖细胞(EPCs)功能障碍的影响及可能机制。方法:将不同浓度黄连素(100,200,400μmol·L^-1)分别加入EPCs中孵育进行活性检测,选取最佳作用浓度。将最佳浓度的黄连素与高糖(40 mmol·L^-1)共孵育,诱导小鼠骨髓来源EPCs损伤,采用CCK-8比色法检测不同时间点(24 h和48 h)的细胞活性;通过小管形成、迁移和黏附能力检测EPCs功能;检测一氧化氮(NO)含量和超氧化物歧化酶(SOD)活性。结果:筛选出200μmol·L^-1为黄连素最佳作用浓度;黄连素显著提高高糖诱导的EPCs细胞活性(P<0.05);显著改善EPCs的小管形成、迁移和黏附功能(P<0.05);显著上调高糖诱导的EPCs的NO含量和SOD活性(P<0.05)。结论:黄连素可能通过调节高糖诱导的NO和SOD水平改善EPCs功能。
Objective:To explore the effect of berberine on endothelial progenitor cells(EPCs)induced by high glucose and its possible mechanism.Methods:Different concentrations of berberine(100,200,400μmol·L^-1)induced EPCs were respectively performed by activity tests to select the appropriate concentration.The optimal concentration of berberine and high glucose(40 mmol·L^-1)were incubated together to induce damage to EPCs derived from bone marrow in mice.The cell viability at different time points(24 h and 48 h)was detected by CCK-8,the EPCs function was detected through tubule formation,migration and adhesion,and nitric oxide(NO)content and superoxide dismutase(SOD)activity were detected.Results:The concentration of 200μmol·L^-1 was selected as the optimal concentration of berberine.Berberine significantly increased the cell viability of EPCs induced by high glucose(P<0.05),significantly improved the tubule formation,migration and adhesion functions of EPCs(P<0.05),significantly up-regulated the NO content and SOD activity of EPCs induced by high glucose(P<0.05).Conclusion:Acarbose might regulate NO level and SOD activity to improve EPCs function induced by high glucose.
