晚期糖基化终末产物对人脐静脉内皮细胞分泌NO功能的影响 被引量:4

摘要 目的探讨晚期糖基化终末产物(AGEs)对人脐静脉内皮细胞分泌一氧化氮(NO)功能的影响。方法制备AGEs修饰的人血白蛋白(AGEs-HSA)。将体外培养的人脐静脉内皮细胞随机分为人血白蛋白(HSA)组和AGEs-HSA组,分别用不同质量浓度的AGEs-HSA、HSA刺激细胞24h。然后用酶联免疫法分别测定细胞培养上清液中的NO水平、一氧化氮合酶(NOS)活性、超氧化物歧化酶(SOD)活性和丙二醛(MDA)水平。结果与HSA组相比,AGEs-HSA组在质量浓度为100、200、400mg/L时能够明显抑制NO的产生和NOS的活性,并呈浓度依赖性(P<0.05)。与HSA组相比,AGEs-HSA组在质量浓度为25、50、100、200、400mg/L时均能够明显抑制SOD活性和促进MDA水平增高,并呈浓度依赖性(P<0.05)。AGEs-HSA组在400mg/L时,NO水平和NOS活性、SOD活性呈正相关(r分别为0.93、0.92),与MDA水平呈负相关(r=-0.83),NOS的活性与SOD活性呈正相关(r=0.90),与MDA水平呈负相关(r=0.87)。结论 AGEs能抑制人脐静脉内皮细胞NO的分泌,其机制之一可能是介导内皮细胞发生氧化损伤致使NOS活性降低进而导致内皮细胞合成NO减少。
