黄连素对血糖波动诱导的内皮损伤的保护机制

摘要 目的研究黄连素是否通过Toll样受体的炎症信号通路改善血糖波动诱导的内皮损伤。方法2022年7月—2023年1月在辽宁沈阳选取雄性Wistar大鼠20只,采用随机数表法分为对照组、波动血糖组、黄连素高剂量组[100 mg/(kg·d)]、黄连素低剂量组[25 mg/(kg·d)],每组5只。应用Ellisa法检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、Toll样受体4(toll-like receptor 4,TLR-4)和天冬氨酸特异性半胱氨酸蛋白酶-3(cysteinyl aspartate-specific proteinase-3,Caspase-3)。结果波动血糖组的TNF-α(30.05±10.65)pg/mL、TLR-4(123.88±19.72)ng/mL、Caspase-3(4.5±0.99)pmol/L表达高于对照组的TNF-α(18.06±4.86)pg/mL、TLR-4(86.28±19.85)ng/mL、Caspase-3(3.36±0.63)pmol/L,差异有统计学意义(P<0.05)。应用高剂量黄连素后的TNF-α(19.28±5.75)pg/mL、TLR-4(89.19±20.02)ng/mL、Caspase-3(3.62±0.51)pmol/L和应用低剂量黄连素后的TNF-α(20.22±6.22)pg/mL、TLR-4(91.41±29.50)ng/mL、Caspase-3(3.63±0.15)pmol/L和波动血糖组比较,差异有统计学意义(P<0.05)。结论血糖波动可增强主动脉内皮细胞中的炎症反应,启动凋亡,应用黄连素后可改善。 Objective To investigate whether berberine can ameliorate blood glucose fluctuation-induced endothelial injury through the inflammatory signaling pathway of Toll-like receptors.Methods 20 male Wistar rats were selected from July 2022 to January 2023 in Shenyang,Liaoning province,and divided into control group,fluctuating blood glucose group,berberine high-dose group[100 mg/(kg·d)]and berberine low-dose group[25 mg/(kg·d)]by random number table method,with 5 rats in each group.Application of Ellisa method for detecting tumor necrosis factor-α(tuber crossing factor-α,TNF-α)、toll like receptor 4(TLR-4)and aspartate specific protease-3(Caspase-3).Results The expressions of TNF-α(30.05±10.65)pg/mL,TLR-4(123.88±19.72)ng/mL and Caspase-3(4.5±0.99)pmol/L in the fluctuated blood glucose group were higher than those in the control group(18.06±4.86)pg/mL and TLR-4(86.28±19.85)ng/mL,Caspase-3(3.36±0.63)pmol/L,and the differences were statistically significant(P<0.05).TNF-α(19.28±5.75)pg/mL,TLR-4(89.19±20.02)ng/mL,and Caspase-3(3.62±0.51)pmol/L after the application of berberine high-dose group,and TNF-α(20.22±6.22)pg/mL,TLR-4(91.41±29.50)ng/mL,and Caspase-3(3.63±0.15)pmol/L after the application of berberine low-dose group,were compared with those in fluctuating blood glucose group,and the differences were statistically significant(P<0.05).Conclusion Blood glucose fluctuation can enhance the inflammatory response and initiate apoptosis in aortic endothelial cells,which can be improved by the application of berberine.
