摘要
目的:建立快速测定比格犬血浆中地佐辛和右美托咪定浓度的超高效液相色谱-质谱(UPLC-MS/MS)法,并用于比格犬体内的药动学研究。方法:血浆采用乙腈沉淀方法处理后检测,以地西泮为内标,色谱柱为Waters Acquity UPLC BEH C18柱(50 mm×2.1 mm,1.7μm),流动相为乙腈和0.1%甲酸,梯度洗脱,流速为0.4 ml·min^-1,柱温为40℃。质谱采用电喷雾(ESI)离子源正离子模式多反应监测(MRM),地佐辛、右美托咪定和内标地西泮的离子对分别为m/z 246.20→147.00、m/z 201.10→94.90、m/z 285.00→154.00。结果:地佐辛在1~200μg·L^-1、右美托咪定0.1~20μg·L^-1范围内具有良好的线性,定量下限分别为1μg·L^-1和0.1μg·L^-1。日内和日间精密度的RSD均在15%以内,绝对回收率在80%以上。结论:本方法快速、简便、准确,可应用于比格犬体内药动学及药物相互作用研究。
Objective:To establish an efficient,sensitive and rapid UPLC-MS/MS method for the simultaneous determination of dezocine(DEZ)and dexmedetomidine(DEX)in beagle’s plasma.Methods:After the plasma was precipitated by acetonitrile,it was detected on an Acquity UPLC BEH C18 column using acetonitrile-formic acid as the mobile phase in a gradient elution mode,and the flow rate was 0.4 ml·min-1.In a positive ion mode,DEZ,DEX and diazepam(IS)were monitored by multiple reactions monitoring(MRM)as the following mass transition pairs:m/z 246.20→147.00 for DEZ,m/z 201.10→94.90 for DEX,and m/z 285.00→154.00 for IS.Results:DEZ and DEX had good linearity within the range of 1-200μg·L^-1 and 0.1-20μg·L^-1,respectively,and the lower limit of quantification was 1μg·L^-1 and 0.1μg·L^-1,respectively.The RSDs of intra-and inter-day precision were within 15%,and the absolute recovery was over 80%.Conclusion:The method is an efficient tool for the simultaneous determination of DEZ and DEX,which may be successfully applied in pharmacokinetic study and DDIs in beagles.
