摘要
目的建立同时测黑骨藤提取物中3-O-咖啡酰基奎宁酸、4-O-咖啡酰基奎宁酸和5-O-咖啡酰基奎宁酸的HPLC-MS/MS分析方法,研究黑骨藤提取物中3种活性成分在佐剂性关节炎(adjuvant arthritis,AA)大鼠体内的药代动力学特征。方法采用弗氏完全佐剂制备AA大鼠模型,于造模d21口服黑骨藤提取物,HPLC-MS/MS法测定AA模型大鼠血浆中3种成分的浓度,葛根素做内标。采用WinNonlin 6.4软件计算药动学参数。结果3种成分的提取回收率和基质效应为81.19%~111.94%,日内和日间精密度的RSD(%)均小于13%,稳定性及线性关系良好,满足生物样品测定要求;3-O-咖啡酰基奎宁酸、4-O-咖啡酰基奎宁酸和5-O-咖啡酰基奎宁酸的Cmax分别为(144.60±23.13)、(72.07±7.02)、(71.31±23.14)μg·L^-1,Tmax分别为(1.00±0.00)、(1.00±0.00)、(1.33±0.29)h。结论建立的HPLC-MS/MS法专属性强,灵敏度高,快速准确,并成功应用于黑骨藤的药动学研究。
Aim To establish a HPLC-MS/MS method for quantifying 3-O-caffeoylquinic acid,4-O-caffeoylquinic acid and 5-O-caffeoylquinic acid and to investigate the pharmacokinetics of three active components in Periploca forrestii extract in the plasma.Methods The adjuvant-induced arthritis(AA)rats were induced by Freund′s complete adjuvant and administered with Periploca forrestii extract,the concentrations of three active components in plasma were determined by HPLC-MS/MS,pharmacokinetic parameters were estimated using the WinNonlin 6.4 software and the data were analyzed by SPSS 22.0 statistical software.Results The extraction recovery and matrix effect of three components ranged from 81.19%to 111.94%,the RSD(%)of intra-day and inter-day precision was less than 13%,and the stability and linear relationship were good,which met the requirements of biological sample determination.The parameters of Cmax(h)in three index components were 144.60±23.13,72.07±7.02,71.31±23.14μg·L^-1,respectively,and Tmax(h)in three index components was 1.00±0.00,1.00±0.00,1.33±0.29,respectively.Conclusion The HPLC-MS/MS method with its strong specificity,high sensitivity,accuracy and rapidness,has been successfully applied to the pharmacokinetic study of the Periploca forrestii extract.
