摘要
本研究采用ACQUITY UPLC^®BEHC18(50 mm×2.1 mm,1.7μm)色谱柱,以0.2%乙酸水溶液(A)-乙腈溶液(B)为流动相进行梯度洗脱,体积流量为0.3 mL/min,检测波长为250 nm,在该色谱条件下测定21批牛膝药材甾酮类成分含量。并关联生物活性项下的ABTS自由基清除率、DPPH自由基清除率和羟基自由基清除率的结果,最后建立用于等级评价的Logistic回归模型。通过该模型,21批的牛膝药材被分成了优良中差四个等级。研究结果表明,基于二分类Logistic回归模型描述牛膝药材等级与影响因素之间的映射关系是可行的,为牛膝药材的质量评价标准的制定提供了新的思路。
The UPLC chromatographic column was acquity ACQUITY UPLCBEHC18(50 mm×2.1 mm,1.7μm),with 0.2%acetic acid-acetonitrile as the mobile phases in gradient solvent system.The detection wavelength was set at 250 nm with flow rate of 0.3 mL/min,and phytoecdysones components were determined in the chromatographic conditions.Then the antioxidant biological activity was reflected through ABTS radical scavenging rate,DPPH radical scavenging rate and hydroxyl radical scavenging rate.Finally,a two-classification logistic regression model was based on correlating biological activity and components.All 21 batches of Achyranthes bidentata were divided into four grades according to this model.The results showed that it was feasible to describe the mapping relationship between the grade and the influencing factors by this model,which could provide a new idea for the evaluation standard of A.bidentata.
