摘要
目的分析结直肠癌患者DNA甲基转移酶1(DNMT1)、组蛋白去乙酰化酶1(HDAC1)的表达及其与临床病理特征和预后的相关性。方法选取本院2015年4月-2017年11月收治的88例结直肠癌患者为研究对象,同时取24例距离肿瘤组织≥5 cm的肠黏膜作为正常组织。观察HDAC1和DNMT1在癌旁正常组织与结直肠癌组织中的表达情况,分析二者在结直肠癌组织中表达与其病理特征及预后的关系。结果结直肠癌组织DNMT1、HDAC1阳性表达率显著高于癌旁正常组织(P<0.01)。DNMT1、HDAC1在结直肠癌组织中的阳性表达率与组织学分化程度呈反比,分化程度越低阳性表达率越高(P<0.05或P<0.01);Dukes分期A、B期和肿瘤未侵及外膜的结直肠癌组织DNMT1、HDAC1阳性表达率均明显低于Dukes分期C、D期和肿瘤侵及外膜的结直肠癌组织(P<0.05或P<0.01);淋巴结有转移者的结直肠癌组织DNMT1、HDAC1阳性表达率也显著高于淋巴结无转移者的结直肠癌组织(P<0.05或P<0.01)。二者表达情况与患者术后2年生存率比较差异无统计学意义(P>0.05)。结论DNMT1与HDAC1表达在结直肠癌的发生、发展及浸润转移等过程中发挥着重要作用,临床可据二者表达情况评估患者病情并指导治疗。
Objective To analyze expression of DNA methyltransferase 1(DNMT1)and histone deacetylase 1(HDAC1)in patients with colorectal cancer(CRC)and their correlation with clinicopathological features and prognosis.Methods 88 patients with CRC treated in our hospital from April 2015 to November 2017 were selected as research subjects,and in meantime,intestinal mucosa of 24 cases located at≥5 cm away from tumor tissue were taken as normal tissues.The expressions of HDAC1 and DNMT1 in adjacent normal tissues and CRC tissues were observed,relationships between their expressions in CRC tissues and pathological features,prognosis of CRC were analyzed.Results The positive expression rates of DNMT1 and HDAC1 in CRC were significantly higher than those of adjacent normal tissues(P<0.01).The positive expression rates of DNMT1 and HDAC1 in CRC were inversely proportional to degree of histological differentiation.The lower degree of differentiation was,higher positive expression rates of DNMT1 and HDAC1 were(P<0.05 or P<0.01).The positive expression rates of DNMT1 and HDAC1 in CRC at Dukes stage A and B and without invading outer membrane were significantly lower than those in CRC at stage C and D and invading outer membrane(P<0.05 or P<0.01).The positive expression rates of DNMT1 and HDAC1 in CRC with lymph node metastasis were also significantly higher than those in CRC without lymph node metastasis(P<0.05 or P<0.01).There was no statistically significant difference between expression of DNMT1 and HDAC and 2-year survival rate of patients after surgery(P>0.05).Conclusion The expression of DNMT1 and HDAC1 play an important role in occurrence,development,invasion and metastasis of CRC.In clinical treatment,the patient's condition can be evaluated and treated according to expression of DNMT1 and HDAC1.
