蒙药药对广枣-肉豆蔻对Wistar乳鼠心肌细胞氧化损伤的保护作用 被引量:10

摘要 目的:探讨蒙药药对广枣-肉豆蔻对Wistar乳鼠心肌细胞氧化损伤的干预作用。方法:取新生Wistar乳鼠心肌细胞原代培养,300μmol/L H2O2诱导建立心肌细胞氧化损伤模型,MTT法观察广枣水提物、广枣10%~95%醇提物、肉豆蔻超临界提取物和广枣-肉豆蔻药对对氧化损伤的心肌细胞和正常心肌细胞的影响,并用qRT-PCR方法检测Bcl-2和Bax的基因表达。结果:诱导损伤模型组细胞生存率较空白组显著降低(P<0.05或P<0.01);广枣水提物无抗氧化损伤活性,不同浓度的广枣醇提物、肉豆蔻挥发油和半固体组及广枣-肉豆蔻药对均能显著增加细胞生存率,且多表现与剂量呈正相关。筛选出70%广枣醇提物、药对广枣-肉豆蔻(70%广枣醇提物∶肉豆蔻挥发油+肉豆蔻半固体成分=1∶1)抗氧化活性最强(P<0.05或P<0.01)。广枣、肉豆蔻、广枣-肉豆蔻药对均能显著调节H2O2诱导的氧化损伤心肌细胞凋亡相关基因Bcl-2、Bax表达,增加Bcl-2/Bax比值(P<0.01),广枣-肉豆蔻药对最强,广枣、肉豆蔻次之。结论:蒙药药对广枣-肉豆蔻能显著降低体外Wistar乳鼠心肌细胞的氧化损伤,抑制心肌细胞凋亡的发生。 Objective: To study the effect of Mongolian common herb pair Choerospondiatis-Nutmeg against to oxidative damage. Methods: Myocardial cells of Wistar neonatal rat cardiomyocytes were acquired and cultured, and 300 μmol/L H2O2 induced myocardial cell oxidative damage. MTT method was used to observe the effects of water, ethanol extract, supercritical extract of Nutmeg and medicine on oxidative injury of cardiomyocytes and normal cardiomyocytes, and the expressions of Bcl-2 and Bax genes were detected by qRT-PCR method.Results: The cell survival rate of induced injury model group was significantly lower than that of control group(P<0.01 or P<0.05). The aqueous extract of Choerospondiatis had no antioxidant activity, ethanol extracts of different concentrations of Choerospondiatis, the essential oil and semisolid component of Nutmeg and Choerospondiatis-Nutmeg pair significantly increased the cell survival rate. The cell vitality are positively correlated with the dose in most cases. Sifting out the 70% ethanol extracts of Choerospondiatis, and Choerospondiatis-Nutmeg pair(70% Choerospondiatis: the essential oil of Nutmeg+ the semisolid of Nutmeg=1∶1) showed stronger antioxidant activity. The genes expression of Bcl-2 and Bax related with apoptosis were significantly regulated by Choerospondiatis, Nutmeg and the pair. The ratio of Bcl-2/Bax was increased(P<0.01). The pair is strongest and Choerospondiatis, Nutmeg second. Conclusion: Mongolian herb pair Choerospondiatis-Nutmeg can reduced the oxidative damage of myocardial cells and inhibit the apoptosis in vitro.
