摘要
目的通过观察蒙药额尔敦-乌日勒预处理对大鼠心肌缺血再灌注损伤(MIRI)细胞凋亡及相关蛋白的影响,探讨额尔敦-乌日勒预处理对MIRI的保护机制。方法将60只健康雄性SD大鼠随机分为6组:假手术组、MIRI模型组、复方丹参滴丸组、额尔敦-乌日勒高、中、低剂量组,每组10只大鼠。给药组分别灌胃给予不同药物预处理。假手术组只穿线不结扎,其余各组大鼠通过结扎冠状动脉左前降支30 min,然后再灌注120 min的方法制作MIRI模型。采用末端标记(tunel)方法检测心肌细胞凋亡率;运用Western Blot技术检测大鼠心肌细胞中Bcl-2,Bax,Caspase-3蛋白表达。结果(1)与假手术组比较,模型组大鼠心肌细胞凋亡率显著上升(P<0.01);与模型组比较,额尔敦-乌日勒能降低MIRI大鼠心肌细胞凋亡率(P<0.01),其中额尔敦-乌日勒中、低剂量组优于高剂量组(P<0.01)。(2)与假手术组比较,模型组大鼠Bcl-2表达显著下调(P<0.01),Bax、Caspase3表达明显上调(P<0.01),Bcl-2/Bax比值下降(P<0.01);与模型组比较,额尔敦-乌日勒能增加MIRI大鼠心肌Bcl-2表达,减少Bax、Caspase3蛋白的表达,提高Bcl-2/Bax比值(P<0.01),其中以额尔敦-乌日勒中剂量组作用较明显(P<0.01)。结论额尔敦-乌日勒可通过抑制心肌细胞凋亡对大鼠MIRI起到保护作用,其抗心肌细胞凋亡的机制可能与通过上调Bcl-2表达,下调Bax、Caspase3的表达,提高Bcl-2/Bax比值有关。
Objective To observe the effects of Eerdun-wurile pretreatment on myocardial ischemic-reperfusion injury(MIRI)cell apoptosis and related proteins,e explored the protective mechanism of the pretreatment of erdun-uzhler on MIRI.Methods Sixty healthy male SD rats were randomly divided into 6 groups:sham operation group,MIRI model group,Compound Danshen Dripping Pill group,Eerdun-wurile high dose group,Eerdun-wurile medium dose group,eerdun-wurile low dose group,10 rats in each group.The administration groups were given different drugs by gavage pretreatment.In the sham operation group,the thread was not ligated,and the remaining rats in each group were subjected to ligate the left anterior descending coronary artery for 30 minutes and then reperfusion for 120 minutes to prepare the MIRI model.Cardiomyocyte apoptosis was detected by the tunel method.We detected Bcl-2,Bax and Caspase-3 Protein expressions in rat cardiomyocytes by Western blot.Results(1)The result of tunel method showed that the apoptosis rate of myocardial cells in MIRI group was significantly higher than that in sham operation group(P<0.01).Compared with the model group,Eerdun-wurile could reduce the myocardial cell apoptosis rate in MIRI rats(P<0.01).Among them,middle and low doses of Eerdun-wurile were better than the high-dose group(P<0.01).(2)The expression of Bcl-2 was significantly decreased in the MIRI group,and the expressions of Bax and Caspase3 were significantly increased(P<0.01),and the Bcl-2/Bax ratio was decreased(P<0.01).Compared with the model group,Eerdun-wurile increased the expression of Bcl-2,decreased the expression sof Bax and Caspase3 and increased the ratio of Bcl-2/Bax in MIRI rats(P<0.01).The effect of middle-dose Eerdun-wurile was more obvious than that of the control group(P<0.01).Conclusion Eerdun-wurile can protect rat MIRI by inhibiting cardiomyocyte apoptosis.Its mechanism of anti-cardiomyocyte apoptosis may be through up-regulation of Bcl-2 expression,down-regulation of expressions of Bax and caspase3,and increase the
