摘要
目的探讨生姜挥发油对脑缺血再灌注损伤大鼠溶酶体-线粒体途径凋亡的影响。方法将大鼠随机分为假手术组、模型组、尼莫地平组(30 mg·kg^-1,阳性对照组)、生姜挥发油高剂量组(0.12 mL·kg^-1)和生姜挥发油低剂量组(0.06 mL·kg^-1)。各组大鼠灌胃给予相应的药物,假手术组和模型组给予等体积的1%的吐温80溶液,每天给药1次,连续给药3 d。末次给药1 h后行手术,用四动脉阻断法复制脑缺血再灌注大鼠模型,再灌注30 min后取大脑;假手术组只分离血管不结扎。用原位末端标记法(TUNEL)检测大鼠脑组织细胞凋亡,蛋白免疫印迹法(Western Blot)测定细胞色素C(Cytochrome C,CytC)、组织蛋白酶B(Cathepsin B,CTSB)和组织蛋白酶D(Cathepsin D,CTSD)的蛋白表达。结果与假手术组比较,模型组皮层中凋亡细胞数明显增加(P<0.01),皮层和海马中细胞色素C、组织蛋白酶B和组织蛋白酶D蛋白表达均明显升高(P<0.01);与模型组比较,生姜挥发油高、低剂量组皮层中凋亡细胞数均明显降低(P<0.01),生姜挥发油高剂量组的皮层和海马中细胞色素C、组织蛋白酶B和组织蛋白酶D蛋白表达均明显降低(P<0.01,P<0.05),生姜挥发油低剂量组的皮层中组织蛋白酶B蛋白表达也明显降低(P<0.05)。结论生姜挥发油可能通过阻断溶酶体-线粒体途径抑制脑缺血再灌注模型大鼠神经细胞的凋亡,从而发挥保护脑损伤的作用。
Objective To investigate the effect of ginger volatile oil on lysosomal-mitochondrial apoptosis in cerebral ischemia-reperfusion(CIR)in rats.Methods The CIR rat model was established by 4 arterial occlusion.Rats were randomly divided into Sham group,CIR model group,nimodipine group(30 mg·kg^-1·d^-1,positive control),ginger volatile oil high dose group(H-dose,0.12 mL·kg^-1·d^-1)and low dose group(L-dose,0.06 mL·kg^-1·d^-1).Rats were treated with nimodipine,ginger or solvent control ig once a day,lasted for 3 days.Surgery was performed at 1 h after drug administration,the vessel of rats in Sham group was separated but not blocked;and then rats were sacrificed after 30 min reperfusion.Terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL)and Western Blot were performed to detect apoptosis of cell in rat brain tissue and protein expression of cytchrome C(CytC),cathepsin B(CTSB),cathepsin D(CTSD)in cortex and hippocampus of rats.Results Compared with Sham group,the number of apoptotic cell was significantly increased in CIR model group(P<0.01),while protein levels of CytC,CTSB and CTSD in cortex and hippocampus of CIR model group were also increased significantly(P<0.01).Compared with CIR model group,apoptotic cell numbers were reduced significantly in H-dose and Ldose groups(P<0.01),the protein levels of CytC,CTSB and CTSD were decreased significantly in H-dose group(P<0.01,P<0.05),while the protein level of CTSB was decreased significantly in L-dose group either(P<0.05).Conclusion Ginger volatile oil is likely to protect CIR injured rats by arresting neuron apoptosis through blocking the lysosomal-mitochondrial pathway.
