摘要
目的检测钕水杨酸硫代脯氨酸配合物对小鼠的急性毒性LD50和致突变性。方法采用经口灌胃小鼠急性毒性试验,设置5个剂量组、阳性对照(环磷酰胺)、阴性对照(食用植物油),根据寇氏法获取LD50;设置低、中、高三个配合物浓度组,观察小鼠骨髓细胞微核率形成情况。结果该配合物LD50为339.4 mg/kg,95%可信限:315.4~365.3 mg/kg,钕水杨酸硫代脯氨酸的PCE微核率与阳性对照组之间显著性差异(P<0.05),配合物浓度为169.72 mg/kg时与阳性组比较微核率小且差异极显著差异(P<0.01)。结论:实验提示钕水杨酸硫代脯氨酸配合物经口灌胃对小鼠具有一定的毒性,微核实验表明在169.72 mg/kg浓度时该配合物对小鼠有致突变作用。
Objective To detect acute toxicity LD50 and mutagenic effect of neodymium salicylate thiproline complex on mice. Methods Acute toxicity test of mice was performed by oral gavage. Five dose groups, positive control(cyclophosphamide) and negative control(vegetable oil) were set. LD50 was obtained by Coleus method. The micronucleus rate of mouse bone marrow cells was observed in the low, medium and high concentration groups. Results The LD50 of the complex was 339.4 mg/kg, and the 95 % confidence limit was 315.4~365.3 mg/kg. At the concentration of 169.72 mg/kg, the micronucleus rate of the positive group was lower and the difference was very significant(P<0.01). Conclusion: The neodymium salicylic acid thiproline complex has certain toxicity to mice by oral gavage, and the micronucleus test shows that the complex has mutagening effect on mice at the concentration of 169.72 mg/kg.
