摘要
采用生物酶解法对包裹在杜仲胶外层的植物组织进行降解以获得原生态杜仲胶丝。以酶解液中的糖含量和酶解后粗胶中的杜仲胶含量为指标,先对生物酶种类及复配酶配比进行筛选,随后用实验因子设计考察了酶解温度、酶解时间、酶解pH值及酶液浓度对酶解效果的影响。结果表明,复配酶的酶解效果要优于单一酶,酶解工艺的最佳条件为:酶解温度50℃,酶解时间4 h,酶解pH值6.0,复配酶液质量浓度4.0 g/L。在此优化条件下,酶解后粗胶原料中的杜仲胶质量分数可达(47.42±1.07)%,大量的杜仲胶丝暴露在植物残渣外层,而且保持了杜仲胶丝的原生状态。
In order to obtain the original ecological gutta-percha,the seed skins of Eucommia ulmoides after seed removal and oil extraction were used as raw materials.With the sugar content in the hydrolysis solution and the gutta-percha content in the crude gutta-percha as the indicators,the enzyme species and ratios were chosen first,and then the effects of the enzymatic hydrolysis temperature,time,pH value and enzyme concentration on the hydrolysis effect were investigated by factor design experiments.The results showed that the hydrolysis effect of complex enzyme was better than that of the single enzyme.The optimum conditions for the enzymatic hydrolysis were:enzymatic hydrolysis temperature 50℃,hydrolysis time 4 h,hydrolysis pH value 6.0,and enzyme concentration 4.0 g/L.Under optimized conditions,the mass fraction of gutta-percha after enzymatic hydrolysis was up to(47.42±1.07)%.A large amount of gutta-percha was exposed to the outer layer of plant residue,and the original ecological gutta-percha was maintained.
