摘要
目的观察木犀草素对卵巢过度刺激综合征(OHSS)模型大鼠炎症因子水平及血管通透性的影响。方法 24只SD大鼠随机分为正常对照组、模型对照组、木犀草素组,每组8只,模型对照组和木犀草素组大鼠皮下注射孕马血清促性腺素(PMSG)和人绒毛膜促性腺素(HCG)建立OHSS模型,正常对照组以等量生理盐水皮下注射;木犀草素组予木犀草素混悬液20 mg/(kg·d)灌胃,正常对照组和模型对照组分别予等量0.5%羧甲基纤维素钠溶液灌胃,连续给药1周;给药结束后,称取卵巢重量,ELISA法检测各组大鼠卵巢组织中白细胞介素-6(IL-6)和血管内皮生长因子(VEGF)的水平,Western blot法检测各组大鼠卵巢组织中血管内皮生长因子受体2(VEGFR-2)的表达。结果模型对照组大鼠卵巢重量明显高于正常对照组(P<0.05),木犀草素组大鼠的卵巢重量显著低于模型对照组(P<0.05);模型对照组大鼠卵巢中IL-6、VEGF、VEGFR-2的表达水平均明显高于正常对照组(P<0.05),而木犀草素组大鼠卵巢中IL-6、VEGF、VEGFR-2的表达较模型对照组明显减少(P<0.05)。结论木犀草素可有效抑制OHSS大鼠卵巢组织的炎症反应及血管通透性。
Objective To observe the effects of luteolin on inflammatory factor level and vascular permeability in rats with ovarian hyperstimulation syndrome(OHSS).Methods Twenty-four SD rats were randomly divided into negative control group(Group NC),positive control group(Group PC)and luteolin treatment group(Group PC+Lut).Subcutaneous injection of pregnant horse serum gonadotropin(PM SG)and human chorionic gonadotropin(HCG)was applied to establish OHSS model in Group PC and Group PC+Lut;while Group NC received equal amount of saline as placebo.The rats in Group PC+Lut received was luteolin by 20mg/(kg·d),while 0.5% sodium carboxymethylcellulose solution as placebo was given in Group NC and Group PC.After treatm ent,the rats were sacrificed and the ovaries were weighed.ELISA was perfomied to test the levels of interleukin-6(IL-6)and vascular endothelial growth factor(VEGF)in ovarian tissues.Western blot was used to examine the protein expression of vascular endothelial growth factor receptor 2(VEGFR-2)in ovarian tissues.Results The ovarian weight of Group PC was significantly higher than that of Group NC(P<0.05),while the ovary weight of Group PC+Lut was significantly lower than that of Group PC(P<0.05).The levels of IL-6 and VEGF by ELISA and the expression levels of VEGFR-2 in ovarian tissues of Group PC were significantly higher than those of Group NC(P<0.05),while the levels of IL-6,VEGF and VEGFR-2 in ovarian tissues of Group PC+Lut were significantly lower than those of Group PC(P<0.05).Conclusion Luteolin effectively inhibits the inflammatory response and vascular permeability of ovarian tissues in OHSS rats.
