高效液相色谱法同时测定甜叶菊中9种甜菊糖苷类化合物 被引量:8

摘要 目的:研究甜叶菊原料中甜菊糖苷类的检测前处理方法,建立一种同时测定甜叶菊中9种糖苷类化合物的高效液相色谱检测方法。方法:采用MGⅡ4.6mm×250mm,5μm色谱柱,以乙腈-磷酸缓冲液溶液(10mM,pH2.6)为流动相进行等度洗脱分离,在紫外波长210nm下进行检测,外标法定量结果:在0.125~4.0mg/mL浓度范围内线性良好,相关系数为0.99996;方法的检出限为0.25%,定量限为0.83%;样品中瑞鲍迪苷A加标回收率为97%~104%。结论:本方法操作简单,分离度高,可同时测定甜叶菊及其提取物中的9种甜菊糖苷组分,可进一步用于甜叶菊的质量控制。 Objective:To study the pretreatment method for detecting steviosides in stevia,a high performance liquid chromatography method for the simultaneous determination of nine steviosides in stevia was established.Methods:Conventional column of Shiseido MGII(4.6mm×250mm,5μm)was used for isocratic elution with acetonitrilephosphate buffer solution(10 mM,pH 2.6)as mobile phase,and detected at UV wavelength of 210 nm.Quantitative results of external standard method:The linearity was good in the concentration range of 0.125~4.0mg/mL,and the correlation coefficient was 0.99996.The detection limit and the quantification limit of the method was 0.25%and 0.83%,respectively.The recoveries of the rebaudioside A was 97%~104%.Conclusion:The method was simple in operation and high in resolution.It could simultaneously determine the nine steviosides in stevia and its extract,which could be further used for quality control of stevia raw materials.
