摘要
目的对2017年采集自重庆地区蚊虫标本携带的病毒进行分离鉴定。方法采用组织细胞培养法进行病毒分离,应用高通量测序技术获得病毒分离物的全基因组序列,采用系统发育法进行种类及分子特征分析。结果库蚊标本来源的病毒阳性分离物CQFD2017能引起白纹伊蚊细胞C6/36病变,但不能引起金黄地鼠肾细胞BHK-21、非洲绿猴肾细胞Vero病变。CQFD2017全基因组序列长度为20893 bp,包含6个开放阅读框。系统进化分析发现,该毒株位于Nidovirales病毒目Mesoniviridae病毒科的Yichang病毒所在的进化分支,与Yichang病毒(NC_040534)的核苷酸一致性为98.6%,ORF1a和ORF1b区氨基酸一致性分别为99.06%和99.15%,因此将该毒株命名为Yichang病毒CQFD2017株。结论2017年重庆市库蚊标本分离的CQFD2017株为Yichang病毒。
To isolate and identify viruses from mosquito samples collected in Chongqing in 2017.Tissue culture was used to isolate the virus,next generation sequencing was used generated the genome of the isolated,and phylogenetic analysis was used to analyze its characteristics.The results showed the isolated CQFD2017 can induce cytopathic effect on Aedes albopictus(C6/36)cell,but not Baby Hamster Kidney(BHK-21)cell and African green monkey kidney(Vero)cell.The whole genome of CQFD2017 was 20893 bp in length,containing six open reading frames(ORFs).Phylogenetic analysis showed that the strain was close related to Yichang virus of family Mesoniviridae,order Nidovirales,with 98.6%nucleic acid identity,and 99.06%and 99.15%amino acid identify of ORF1a and ORF 1b regions to Yichang Virus(NC_040534),the isolate was designated Yichang virus isolated CQFD2017.CQFD2017 strain isolated from Culex mosquitoes in Chongqing,China was identified as Yichang virus.
