摘要
[目的]低温胁迫是影响高粱萌发及幼苗长势的重要因素之一,探讨高粱幼苗生理特性对低温胁迫的响应机制,明确科学的耐冷综合评价方法,为高粱耐冷种质的筛选及耐冷品种选育提供理论依据。[方法]以高粱耐冷品种“冻高粱”和“红壳四号”和冷敏品种“SC407”和“晋303”为试验材料,在2℃条件下低温胁迫处理时间分别为12、24、36 h,研究不同冷害胁迫条件对高粱幼苗MDA含量、SOD活性、POD活性及Pro含量的影响,并采用隶属函数法科学评价高粱幼苗的耐冷性。[结果]耐冷和冷敏品种的MDA含量和Pro含量均随着低温胁迫处理时间增加,呈现增高的趋势,冷敏品种的MDA含量增加幅度显著高于耐冷品种(P<0.05),其上升平均幅度变化范围在58.9%~158.2%,而耐冷品种各处理的脯氨酸含量值均高于冷敏品种,其变化范围在18.3%~75.2%;高粱幼苗SOD活性、POD活性随低温处理时间呈现先升高后降低的变化规律,各低温处理的酶活性均高于对照,耐冷品种的两种酶活性平均增加幅度分别为53.6%、43.5%,显著高于冷敏品种的酶活性平均增加幅度(P<0.05),其值分别为31.0%、37.3%;12 h低温胁迫高粱生理指标隶属函数平均值耐冷综合评价U值排序为:SC407<晋303<红壳四号<冻高粱,其结果与生产实际相吻合。[结论]丙二醛含量、脯氨酸含量、过氧化物酶活性和超氧化物歧化酶活性4个生理指标可作为高粱耐冷性的鉴定指标,结合隶属函数法对高粱苗期进行耐冷综合评价科学可行,可以为高粱品种耐冷性鉴定提供参考。
[Objective]The objective of this study was to provide an evaluation method for the screening and breeding of sorghum cold-resistant.[Method]Using the cold-resistant and cold-sensitive Sorghum seedlings as test material,the effects of cold stress on the MDA content,proline content,and SOD and POD activities of Sorghum seedlings were investigated,and the cold tolerance of sorghum varieties was comprehensively evaluated by membership function method.[Result]With the prolongation of low temperature stress time,the MDA and proline contents of both varieties were increased,and the MDA content of cold-sensitive strains was significantly higher than that of cold-resistant strain with the average increased percentage ranged between 58.9%and 158.2%.However,the proline content of cold-resistant strain was constantly higher than that of cold-sensitive strain with the increased percentage ranged between 18.3%and 75.2%.The activities of SOD and POD in leaves were boosted at the beginning and then decreased.The activities of SOD and POD in cold-tolerant lines were steadily higher than that of the control,and the average increases of the two enzyme activities of the cold-tolerant lines were ranged between 53.6%and 43.5%,that were significantly higher than the cold-sensitive varieties,which possessed with the increased values between 31.0%and 37.3%.The results of 12-hour cold tolerance comprehensive evaluation of sorghum physiological index membership function were consistent with the production practice.U value ranking was determined as SC407<Jin303<Red Shell No.4<Frozen Sorghum.[Conclusion]It was feasible to make comprehensive evaluation of cold tolerance of sorghum lines by membership function method and the ontcomes can provide a reference for the identification of sorghum cold tolerance varieties.
