摘要 试验建立HPLC同时测定博落回中不同生长时期不同部位中金黄紫堇碱、碎叶紫堇碱、四氢非洲防己碱含量的方法,并对博落回不同生长时期不同部位中这3种生物碱的含量进行了比较与分析。色谱柱Agilent 5 TC-C18(2)、柱温35℃、波长286 nm、流速0.8 mL/min、进量10μL,流动相磷酸水(A)和磷酸乙腈(B),洗脱条件:0~5 min 5%~15%B,5~25 min 15%~20%B,25~32 min 20%~25%B,32~40 min 25%~40%B,40%~50 min 40%~90%B。结果表明,在上述试验条件下金黄紫堇碱、碎叶紫堇碱与四氢非洲防己碱分离度良好,且在各自的线性范围内线性关系良好,精密度、稳定性、重复性、加样回收率的RSD均在3%以内。金黄紫堇碱在博落回花果期的果含量最高,碎叶紫堇碱在博落回果期叶中含量最高,四氢非洲防己碱则在花期叶中含量最高。该方法操作简便、重复性高,可用于博落回中3种生物碱含量的测定,为博落回作为饲料添加剂的充分开发与利用提供参考。 A HPLC method was established for the simultaneous determination of the contents of scoulerin,cheilanthifoline and tetratydrolumbamine in different parts of different growth periods in Macleaya,and the three alkaloids in different parts of in different growth periods.The content was compared and analyzed.Column was Agilent 5 TC-C18(2),column temperature was 35℃,wavelength was 286 nm,flow rate was 0.8 mL/min,injection was 10μL,mobile phase were phosphate water(A)and acetonitrile phosphate(B),and elution conditions were 0~5min 5%~15%B,5~25 min 15%~20%B,25~32 min 20%~25%B,32~40 min 25%~40%B,40~50 min 40%~90%B.The results showed that the separation of scoulerin,cheilanthifoline and tetratydrolumbamine was good,and the linear relationship between them was good within their respective linear range.The RSD of precision,stability,repeatability and sample recovery were all within 3%.The content of scoulerin was the highest in the fruit stage,cheilanthifoline was the highest in the fruit stage,while tetratydrolumbamine was the highest in the flower stage.The method is simple and reproducible,and can be used for the determination of three alkaloids in Macleaya,providing theoretical basis for the full development and utilization of Macleaya.
