内部沸腾法强化提取血根碱的工艺研究 被引量:10

摘要 试验采用内部沸腾法提取技术,利用响应面分析方法对提取博落回果荚中血根碱的提取工艺进行优化。通过高效液相色谱法测定血根碱的含量,检测波长为271 nm,流动相为乙腈-0.1%磷酸。首先通过单因素试验筛选出血根碱的内部沸腾提取的3个主要因素:解吸剂浓度、硫酸浓度和提取温度;再通过响应面法考察内部沸腾提取法的工艺条件。通过响应面分析确定最佳工艺参数为:解吸剂浓度53%、提取温度82℃、硫酸浓度0.24 mol/L。在最佳条件下,通过内部沸腾法提取得到的血根碱的提取率为9.10 mg/g。内部沸腾法具有省时、高效的优点,适合博落回果荚中血根碱的提取。 The inner ebullition extraction technology was used to optimize the extraction process of extracting sanguinarine from Macleaya cordata by response surface analysis.The content of sanguinarine was determined by UHPLC.The detection wavelength was 271 nm and the acetonitrile-0.1%phosphoric acid was used mobile phase.Firstly,the three main factors of inner ebullition extraction of sanguinarine were screened by single-factor experiments based on the sulfuric acid concentration,desorbent concentration and extraction temperature,and then the process conditions of inner ebullition extraction method were investigated by response surface method.The optimal process parameters were determined by response surface analysis.The desorbent concentration was 53%,the extraction temperature was 82℃,and the sulfuric acid concentration was 0.24 mol/L.Under the conditions,the extraction rate of sanguinarine obtained by inner ebullition method was 9.10 mg/g.The inner ebullition method has the advantages of time saving and high efficiency,and is suitable for the extraction of sanguinarine from Macleaya cordata.
