摘要
目的:探索不同中医证候下的结直肠癌患者的肠道菌群特征。方法:收集首发结直肠癌患者粪便,根据中医证候量表分为脾虚湿阻证、气滞湿阻证、气滞血瘀证、气血亏虚、脾虚气滞人群,利用Illumina Mi Seq技术对各组粪便样本DNA测序。予Alpha多样性指数、PCoA分析、heatmap聚类图、LDA评分挖掘属水平的差异肠道菌群种类。结果:与其他证候相比,脾虚气滞证的Alpha多样性明显降低。各证型的微生物在PCoA分析、heatmap聚类图及LDA评分上显示在属水平上存在差异。UPGMA分析示脾虚湿阻与气血亏虚证候患者的肠道菌群存在关联,脾虚气滞与气滞湿阻患者的肠道菌群存在关联。而气滞血瘀、脾虚湿阻及气血亏虚菌群的LDA评分下多样性较为突出,且有相应证候关联菌群相富集。结论:结直肠癌不同中医证候的肠道菌群存在明显的差异,丰富了中医证候学说的物质基础。
Objective:To explore the characteristics of fecal microbiota in colorectal cancer patients with different Chinese medical syndrome.Methods:The feces of the patients who were first diagnosed as colorectal cancer were collected and divided into spleen deficiency and dampness syndrome,qi stagnation and blood stasis syndrome,deficiency of qi and blood syndrome,spleen deficiency and qi stagnation syndrome and retention of dampness and stagnation of qi syndrome according to the TCM syndrome scale.Illumina Mi Sequence technique was performed to detect the DNA sequence of microbiota as well as Alpha diversity index,principal coordinate(PCoA)analysis,heatmap cluster map,LDA score were prepared to analyze the microbiota.Results:Colorectal cancer patients with spleen deficiency and qi stagnation showed lower diversity than other syndromes,moreover,microbiota from each syndrome showed the difference in PCoA,heatmap cluster map and LDA score in genus level.And UPGMA analysis showed microbiota from spleen deficiency and dampness syndrome correlated with deficiency of qi and blood syndrome as well as retention of dampness and stagnation of qi syndrome was associated with spleen deficiency and qi stagnation syndrome.LDA score in qi stagnation and blood stasis syndrome,spleen deficiency and dampness syndrome and deficiency of qi and blood deficiency of qi and blood showed much more diversity,which may be the biomarker in each syndrome.Conclusion:There has been much more difference of fecal microbiota in colorectal cancer patients with different Chinese medical syndrome,which would enrich the theory of Chinese medical syndrome.
