摘要
目的:探讨马钱子碱对人肾癌细胞ACHN的抑制作用及相关机制。方法:以人肾癌细胞ACHN为研究对象,分别于24 h、48 h、72 h用不同浓度(0μg/mL、100μg/mL、200μg/mL、400μg/mL)的马钱子碱处理,通过MTT法检测细胞增殖抑制情况,流式细胞术检测细胞凋亡情况,Western免疫印迹法检测相关蛋白的表达水平。结果:马钱子碱可抑制人肾癌细胞ACHN增殖,在一定时间、一定浓度范围内呈时间、浓度依赖性;马钱子碱可促进人肾癌细胞ACHN凋亡,在一定时间、一定浓度范围内呈时间、浓度依赖性;随着药物浓度的增加,人肾癌细胞ACHN内DKK1蛋白表达量逐渐增加,β-catenin、MMP-7和C-myc蛋白表达量逐渐降低。与空白对照组比较,差异有统计学意义(P<0.05)。结论:马钱子碱可能通过抑制Wnt/β-catenin信号通路诱导人肾癌细胞ACHN凋亡,从而抑制肾癌细胞的增殖。
Objective:To investigate the inhibitory effect of brucine on human renal cancer cells ACHN and its related mechanism. Methods:Human renal cancer cells ACHN were used as the research object,and were respectively handled with different concentrations(0 μg/mL,100 μg/mL,200 μg/mL,400 μg/mL)of brucine for 24 h,48 h and 72 h. Then cell proliferation was detected by MTT method,cell apoptosis was detected by flow cytometry and Western blot was used to detect the expression level of related proteins.Results:Brucine can inhibit the proliferation of human renal cancer cells ACHN in time-dependent and concentration-dependent manner within a certain time and concentration range;brucine can promote the apoptosis of human renal cancer cells ACHN in concentration-dependent manner within a certain concentration range. With the increase of drug concentration,the expression of DKK1 protein in human renal cancer cells ACHN was gradually increased,and the expressions of β-catenin,MMP-7 and C-myc proteins were gradually decreased(P<0.05).Conclusion:Brucine can induce apoptosis of human renal cancer cells ACHN by inhibiting the Wnt/β-catenin signaling pathway,so as to inhibit the proliferation of renal cancer cells.
