摘要
在铃兰组织培养过程中,外植体通常会发生褐变,这对铃兰组织培养物的正常生长和增殖具有负面影响。分别通过暗培养以及在培养基中添加不同浓度的聚乙烯吡咯烷酮(PVP)、硫代硫酸钠(Na 2S 2O 3)、抗坏血酸(VC)、活性炭(AC)4种抗褐变剂的方法,对铃兰组织培养中发生的褐变进行了防治研究。结果表明,接种后进行暗培养处理以及在培养基中添加抗褐变剂均可显著减少外植体褐变的生成,其中暗培养10 d的效果最佳,抗褐变剂的抑褐效果为VC(2.0 g/L)>AC(0.5 g/L)>PVP(2.0 g/L)>Na 2S 2O 3(0.5 g/L),2.0 g/L的VC的抑褐效果最好,其褐变率为25.33%。
During the tissue culture of Convallaria majalis L.,the explants usually undergo browning,which has a negative impact on the normal growth and proliferation of Convallaria majalis L.tissue culture.Therefore,in this experiment,dark culture and different concentrations of polyvinylpyrrolidone(PVP),sodium thiosulfate(Na 2S 2O 3),ascorbic acid(VC),activated carbon(AC)four anti-browning agents were added to the medium.The browning that occurred in the tissue culture of Convallaria majalis L.was studied.The results showed that the dark culture treatment after inoculation and the addition of anti-browning agent in the medium could significantly reduce the browning of explants.The effect of dark culture for 10 days was the best,and the browning effect of anti-browning agent was VC.(2.0g/L)>AC(0.5 g/L)>PVP(2.0 g/L)>Na 2S 2O 3(0.5 g/L),2.0g/L VC has the best browning effect,and its browning rate is 25.33%.
