摘要 目的探究雌激素对脑缺血再灌注损伤大鼠自由基代谢及神经保护作用。方法将30只健康雄性SD大鼠随机分为假手术组(S组)、缺血再灌注组(I/R组)和雌激素组(E组),每组各10只。S组仅分离动脉血管,不结扎,I/R组和E组建立右侧大脑中动脉闭塞局灶性脑缺血再灌注模型,E组造模前腹腔注射雌激素,测定脑缺血再灌注后脑组织丙二醛(MDA)、过氧化氢酶(CAT)、过氧化物歧化酶(SOD)含量及海马组织肿瘤坏死因子-α(TNF-α)、单核细胞趋化因子(MCP-1)及脑源性神经营养因子(BDNF)蛋白表达。结果与S组比较,I/R组大鼠脑组织神经细胞固缩破裂、排列杂乱,存在空泡样变,有炎性细胞浸润,且脑组织SOD、CAT、BDNF表达下调,MDA、TNF-α和MCP-1水平上调,差异有统计学意义(P<0.05)。E组神经细胞排列有序、结构清晰、形态基本保持完整;E组SOD、CAT高于I/R组,MDA、TNF-α和MCP-1表达下调,BDNF表达上调,差异有统计学意义(P<0.05)。结论雌激素能改善脑缺血再灌注中氧化应激造成的损伤,通过减少MDA释放,增加SOD、CAT水平,下调TNF-α、MCP-1,上调BDNF表达发挥抗炎及保护神经元作用。 Objective To investigate the effects of estrogen on free radical metabolism and neuroprotection in rats with cerebral ischemia reperfusion injury.Methods Thirty healthy male SD rats were randomly divided into sham operation group(Group S),ischemia-reperfusion group(Group I/R)and estrogen group(Group E),with 10 rats in each group.Group S separation arteries,only not ligation,Group I/R and E set up the right focal brain lack of middle cerebral artery occlusion reperfusion model,make model by intraperitoneal injection of former estrogen Group E,determination of cerebral ischemia reperfusion after brain tissue malondialdehyde(MDA),catalase(CAT),superoxide dismutase(SOD)content and the hippocampal tissue tumor necrosis factor alpha(TNF-α),monocyte chemotactic factor(MCP-1)and brain derived neurotrophic factor(BDNF)protein expression.Results Compared with group S,nerve cells in the brain of rats in Group I/R were collapsed and arranged in disorder,with vacuol-like changes and inflammatory cell infiltration,and the expressions of SOD,CAT and BDNF in the brain were down-regulated,and the levels of MDA,TNF-αand MCP-1 were up-regulated,with statistically significant differences(P<0.05).Nerve cells in group E were arranged orderly,with clear structure and basically intact morphology.Compared with Group I/R,the expression of SOD,CAT in Group E were up-regulated,MDA,TNF-αand MCP-1 were down-regulated,while BDNF was up-regulated(P<0.05).Conclusion Estrogen can improve the injury caused by oxidative stress in cerebral ischemia and reperfusion,and play an anti-inflammatory role by reducing the release of MDA,increasing the level of SOD and CAT,and down-regulating the expression of TNF-αand MCP-1 in brain tissue.
