左旋龙脑与左旋樟脑配伍对永久性局灶性中动脉阻断脑缺血模型大鼠的脑保护作用 被引量:1

Brain protective effect of L-borneol and L-camphor on permanent focal middle cerebral artery occlusion rat model

摘要 目的:探究左旋龙脑与左旋樟脑不同配比(以下统称为L-Z x∶y)对永久性局灶性中动脉阻断脑缺血(permanent middle cerebral artery occlusion,pMCAO)大鼠模型的脑保护作用。方法:将大鼠随机分为假手术组、溶剂+模型组、尼莫地平组及L-Z 1∶0、9∶1、8∶2、7∶3、6∶4、 5∶5、0∶1组,共10组。连续预防性给药3 d,末次给药30 min后,采用线栓法建立pMCAO模型,记录苏醒时间;根据Longa、mNss评分法对大鼠术后24 h和96 h神经功能进行评价,后麻醉处死取大脑;TTC染色检测大鼠脑梗死面积,HE染色观察大鼠缺血侧皮层、海马CA1区的病理学形态。结果:与假手术组相比,溶剂+模型组在苏醒时间、神经功能评分、脑梗死面积以及缺血侧皮层和海马CA1区的病理变化方面均显著增加。与溶剂+模型组比较,L-Z 7∶3组极显著延长了大鼠的苏醒时间(P<0.01);L-Z 6∶4组显著降低大鼠Longa评分(P<0.05);L-Z 8∶2、L-Z 7∶3、L-Z 0∶1组显著降低大鼠mNss评分(P<0.05),L-Z 6∶4组极显著降低大鼠mNss评分(P<0.01);L-Z 1∶0组和尼莫地平组显著降低了大鼠的脑梗死面积(P<0.05),L-Z 6∶4组极显著降低了大鼠脑梗死面积(P<0.01);病理形态学显示尼莫地平和L-Z 6∶4组能减缓局部缺血所致的脑组织坏死。结论:左旋龙脑和左旋樟脑组改善了pMCAO模型大鼠的神经功能损伤和脑梗死面积,对pMCAO模型大鼠具有一定的脑保护作用,其中L-Z 6∶4组作用最强。 OBJECTIVE To explore the cerebral protective effect of different ratios of L-bornol to L-camphor(hereinafter collectively referred to as L-Z x∶y) on permanent middle cerebral artery occlusion(pMCAO) rat model.METHODS Male SD rats were randomly divided into sham operation group,solvent+model group,nimodipine group,and L-Z 1∶0,9∶1,8∶2,7∶3,6∶4,5∶5 and 0∶1 ratio groups,and there were 10 groups in total.Continuous prophylactic administration was performed for 3 days,and 30 minutes after the last administration,a pMCAO model was established by the thread embolization method,and the intraoperative awakening time was recorded.According to the Longa and mNss scoring methods,the neurological function of rats was evaluated at 24 and 96 hours after surgery,followed by anesthesia and sampling.TTC staining was used to detect the area of cerebral infarction in rats,while HE staining was used to observe the pathological morphology of the ischemic cortex and hippocampal CA1 area in rats.RESULTS Compared with the sham operation group,the solvent+model group showed a significant increase in recovery time,neurological deficit score,and cerebral infarction area,as well as significant pathological changes in the ischemic cortex and hippocampal CA1 area.Compared with the solvent+model group,the L-Z 7∶3 ratio group significantly prolonged the awakening time of rats after anesthesia(P<0.01);the L-Z 6∶4 ratio group significantly reduced the Longa score of rats(P<0.05);the L-Z 8∶2 ratio group,L-Z 7∶3 ratio group,and L-Z 0∶1 ratio group significantly reduced the mNss score of rats(P<0.05),while the L-Z 6∶4 ratio group significantly reduced the mNss score of rats(P<0.01);the L-Z 1∶0 ratio group and nimodipine group significantly reduced the cerebral infarction area of pMCAO model rats(P<0.05),while the L-Z 6∶4 ratio group significantly reduced the cerebral infarction area of rats(P<0.01).Pathological morphology indicated that the combination of nimodipine and L-Z 6∶4 ratio group could slow down brain tissue
