摘要
目的 探讨三氧化二砷 (As2 O3)对卵巢癌细胞株体外生长的影响。方法 以As2 O3处理卵巢癌细胞株SKOV3,采用四甲基偶氮唑蓝 (MTT)比色法 ,测定As2 O3对SKOV3生长的抑制作用 ,并绘制剂量—效应曲线 ;通过透射电镜观察细胞超微结构的变化 ;应用流式细胞仪分析DNA含量和细胞周期并测定凋亡特征酶caspase 3的活性变化。结果 0 5~ 4 .0 μmol L三氧化二砷明显抑制SKOV3细胞的增殖 ,并呈剂量和时间依赖性 ;透射电镜观察可看到较为典型的细胞凋亡的形态学改变 :细胞核固缩 ,染色质凝集 ,呈新月状紧贴于核膜周边 ,凋亡小体形成等 ;流式细胞仪检测出现DNA含量小于G1 峰的亚二倍体凋亡峰 ,同时出现G2 M期阻滞 ;SKOV3在As2 O3作用下 ,caspase 3是实现凋亡的效应因子 ,它的激活必然导致凋亡细胞最终的各种表型变化。结论 As2 O3对卵巢癌细胞株SKOV3的生长有明显的抑制作用 ,诱导凋亡是其主要作用机制之一。
Objective To explore the effect of arsenic trioxide(As 2O 3) to ovarian cancer cell line SKOV 3.Methods Methyl thiazolyl tetrazolium(MTT) assay was used to establish the dose effect curves.The morphological changes of the cell were observed by transmission electron microscope.The change of cell cycle was analyzed by flow cytometry.The activity of caspase 3 was studied by flurometric immunosorbent enzyme assay(FIENA).Results The inhibition of As 2O 3 to the growth of SKOV 3 was dose dependent.After 24h of exposure to As 2O 3,SKOV 3 cell line presented some morphologic features of apoptosis,including cell shrinkage,chromatin condensation and formation of apoptotic bodies.Some typical subdiploid peaks before G 0/G 1 phase were observed while the cells were arrested in G 2/M phase.As 2O 3 activated caspase 3 and caused apoptosis.Conclusion As 2O 3 can inhibit growth of SKOV 3 and induce apoptosis of it.
