摘要
目的 探讨三氧化二砷 (As2 O3 )诱导膀胱癌BIU 87细胞凋亡作用。方法 应用倒置相差显微镜、透射电子显微镜、自动酶标仪、细胞免疫组化法 ,分别对不同浓度加药组及对照组BIU 87细胞的形态学改变 ,细胞的存活及Bcl 2、Bax蛋白的表达进行观察和测定。结果 生长曲线显示 ,18.8μg ml,37.6 μg ml和 94.0 μg mlAs2 O3 均能抑制膀胱癌BIU 87细胞的生长增殖。透射电镜观察 ,膀胱癌细胞表面的突起减少或脱失 ;部分细胞核染色质聚集 ,边移 ;线粒体膜结构模糊不清。细胞免疫组化法测定Bcl 2、Bax蛋白的表达均有变化 ,与对照组细胞相比 ,18.8μg ml,37.6 μg mlAs2 O3 组Bcl 2和Bax蛋白表达率均升高。结论 三氧化二砷不仅抑制膀胱癌BIU 87细胞增殖 ,而且诱导细胞凋亡。
Objective To study the effect of arsenic trioxide used as chemotheraputic agent on bladder cancer BIU-87 cell.Methods Changes were studied under electron microscopy. The cell growth and proliferation were observed by MTT colorimetric assay and cell-growth curse.The expressions of Bcl-2 and Bax were detected by immunocytochemical method.Results The cell-growth curse revealed that the cell growth was significantly inhibited by the different concentration of arsenic trioxide(18.8μg/ml,37.6μg/ml). Under electron microscope,spines on the surface of the bladder cancer cells decreased. Some chromosome gathered at the margin of cell nuclear. The membrane structures of mitochondrion become obscure. High expression level of Bcl-2 and Bax compared with control were detected in arsenic trioxide-treated cells.Conclusion Arsenic trioxide not only can inhibite proliferation but also induce apoptosis of BIU-87 cells. The induced-apoptosis effect of arsenic trioxide is relative to high expression level of Bax.
