摘要 目的 研究As2 O3 对白血病细胞株NB4 ,K5 6 2 ,HL 6 0的活性氧自由基 (ROS)的影响。方法 细胞内活性氧 (ROS)用DCFH DA标记 ,并用流式细胞仪检测。结果  0 .6 μmol/LAs2 O3 作用 72h能诱导NB4发生 4 4 .9%的凋亡 ,提高其浓度至 2 .7和 8.1μmol/L ,也能诱导K5 6 2和HL 6 0分别发生 5 8.3%和 6 2 .3%凋亡。As2 O3 能显著降低三种细胞内ROS水平 ,且随浓度增加而降低。As2 O3 无论高浓度短时间作用或低浓度长时间作用对细胞内ROS抑制作用是相似的。结论 ROS水平下降在As2 O3 的诱导凋亡作用过程起了重要作用。 Objective Effect of the arsenic trioxide (As 2O 3) on apoptosis and the reactive oxygen species (ROS) levels in three leukemia cell lines NB4, K562 and HL-60 were investigated. Methods ROS were labeled by DCFH-DA and examined by flowcytometry. Results The As 2O 3 with 0.6 μmol/L treated for 72h induced 44.9 % apoptosis only in NB4 cells, however, when concentrations were increased to 2.7 and 8.1 μmol/L respectively, 58.3 % and 62.3 % apoptosis significantly exhibited in K562 and HL-60 cells. Under the above 3 concentrations of As 2O 3, the levels of ROS were sharply decreased in three leukemia cell lines. The effect of higher concentration of As 2O 3 with shorter time was similar to that of lower concentration with longer time. Conclusion The declines of ROS involved in apoptosis induced by As 2O 3.
