摘要
【目的】探讨化瘀祛痰方对高脂血症大鼠心脏功能及脂质代谢的影响。【方法】将40只大鼠随机分为正常组、模型组、化瘀祛痰组和辛伐他汀组,每组10只。正常组给予普通饲料喂养,其余3组给予高脂饲料喂养12周复制高脂血症模型。造模结束后,化瘀祛痰组和辛伐他汀组大鼠分别给予化瘀祛痰方(剂量为16.0 g·kg^-1·d^-1)和辛伐他汀(剂量为3.0 mg·kg^-1·d^-1)灌胃,连续给药30 d。小动物超声影像系统检测大鼠心功能指标变化情况;全自动生化分析仪检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量;蛋白免疫印迹(Western blot)技术检测肝组织中胆固醇酯转移蛋白(CETP)、肝脂酶(HL)蛋白的表达。【结果】与正常组比较,模型组大鼠TG、TC、LDL-C升高,肝组织CETP蛋白表达升高、HL蛋白表达降低,心功能指标异常(P <0.05或P <0.01);与模型组比较,化瘀祛痰组大鼠血脂TG、TC水平降低,肝脏CETP蛋白表达降低、HL蛋白表达升高,心功能改善(P <0.05或P <0.01)。【结论】化瘀祛痰方可降低高脂血症大鼠血脂水平,调节脂质代谢相关蛋白CETP、HL的表达,改善心功能。
Objective To explore the effect of Stasis-resolving and Phlegm-expelling (SP) Recipe on heart function and lipid metabolism in rats with hyperlipidemia. Methods Forty rats were randomly divided into normal group, model group, SP group and simvastatin group, 10 rats in each group. The normal group was fed with ordinary forage, while the other 3 groups were fed with high-fat forage for 12 weeks to induce into hyperlipidemia model. After modeling, SP group and simvastatin group were given intragastric administration of SP Recipe (at dosage of 16.0 g·kg^-1 ·d^-1) and simvastatin (at dosage of 3.0 mg· kg^-1·d^-1), respectively, the treatment lasting 30 continuous days. After medication, the indexes of cardiac function of rats were detected by small animal ultrasonic apparatus , the serum contents of triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and low- density lipoprotein cholesterol (LDL- C) were detected by automatic biochemical analyzer, and the expression levels of cholesterol ester transfer protein (CETP) and hepatic lipase (HL) in liver tissues were detected by Western blotting method. Results Compared with the normal group, the levels of TG, TC, LDL- C were increased in the model group, CETP expression level was increased and HL protein expression level was decreased in liver tissues, and the indexes of cardiac function were abnormal (P < 0.05 or P < 0.01). Compared with the model group, the TG and TC levels were decreased in SP group, CETP expression level was decreased and HL expression level was increased in liver tissues, and the cardiac function indexes were improved (P < 0.05 or P <0.01). Conclusion SP Recipe can decrease the blood lipid levels, regulate the expression of lipid metabolismrelated proteins CETP and HL, and improve the cardiac function of the rats with hyperlipidemia.
