摘要 目的建立测量黄刺玫果中金丝桃苷、芦丁、槲皮素含量的方法,使其得到更好的开发应用。方法建立高效液相色谱方法,选择Inertsil-ODS-SP型号的色谱柱,乙腈(B)-0. 5%磷酸溶液(A)系统作为流动相,洗脱程序为:0-8 min,90%-81%A,10%-19%B; 8-12 min,81%-78%A,19%-22%B; 12-19 min,78%-76%A,22%-24%B; 19-28 min,76%-72%A,24%-28%B; 28-38 min,72%A,28%B; 38-45 min,72%-70%A,28%-30%B; 45-50 min,70%-90%A,30%-10%B; 50-55 min,90%A,10%B。从专属性、精密度、重复性、准确性、线性范围、系统适用性和加样回收率方面对该方法的科学性和可行性进行考察。结果芦丁、金丝桃苷、槲皮素的线性关系相关系数R2分别为0. 999 9,0. 999 9,0. 999 9,线性关系良好。芦丁、金丝桃苷和槲皮素的加样回收率分别为98. 10%,97. 50%和97. 80%。RSD值分别为3. 20%,2. 80%和3. 00%,符合实验要求。结论该方法可行性强,准确性、重复性均符合要求,线性关系良好,适用于测量乙酸乙酯萃取黄刺玫果提取物中芦丁、槲皮素、金丝桃苷的含量。 Objective To establish a method for measuring the contents of hyperoside,rutin and quercetin in the fruit of R. xanthina Lindl. Methods HPLC method was established and performed on Inertsil-ODS-SP type column. The acetonitrile-0. 5% phosphoric acid solution system was used as mobile phase,and the program was 0-8 min,90%-81% A,10%-19 % B;8-12 min,81%-78% A,19%-22% B;12-19 min,78%-76% A,22%-24% B;19-28 min,76%-72% A,24%-28% B;28-38 min,72% A,28% B;38-45 min,72%-70% A,28%-30% B;45-50 min,70%-90% A,30%-10% B;50-55 min,90% A,10%B. The specificity,precision,repeatability,accuracy,linear range,system suitability,and sample recovery of the method were verified. Results The rutin,hyperoside and quercetin showed a good linear relationship( R2= 0. 999 9,0. 999 9,0. 999 9). The recovery rates of rutin,hyperoside and quercetin were 98. 10%,97. 50% and 97. 80%,respectively. The RSD values were 3. 20%,2. 80%,and 3. 00%,respectively,which met the experimental requirements. Conclusion The method has strong feasibility,accuracy and repeatability,and has good linearity. It is suitable for measuring the contents of rutin,quercetin and hyperoside in the extraction of ethyl acetate extract of R. xanthina Lindl.
