摘要
目的:建立白热斯丸质量控制的定性与定量方法。方法:采用薄层色谱法(TLC)对制剂中胡椒碱和槲皮素进行色谱鉴别;采用高效液相色谱法(HPLC)测定制剂中槲皮素和胡椒碱,色谱柱:Shimadzu VP-ODS(250 mm×4.6 mm,5μm);流速:1 mL·min-1;进样量:10μL;柱温:30℃。槲皮素以甲醇-0.2%磷酸溶液(56∶44)为流动相,检测波长:367 nm。胡椒碱以甲醇-水(77∶23)为流动相,检测波长:343 nm。结果:薄层色谱图中能清晰检出槲皮素与胡椒碱;槲皮素在6.25~200μg·mL-1线性关系良好,胡椒碱在6.25~200μg·mL-1线性关系良好(r分别为0.9995、0.9999);槲皮素与胡椒碱平均回收率分别为95.7%和104.8%。结论:本方法专属性强、快速、简便,可作为白热斯丸的质量控制。
Objective:To establish a qualitative and quantitative method for quality control of Baras pill.Methods:TLC was used to identify piperine and quercetin.Quercetin and piperine in the preparation were determined by HPLC.The determination was performed on Shimadzu VP-ODS(250 mm×4.6 mm,5μm)column under temperature of 30℃with mobile phase of methanol-0.2%phosphoric acid solution(56∶44)for quercetin,the flow rate was 1 mL·min-1,and the sample amount was 10μL.The detection wavelength was 367 nm.Piperine was detected at 343 nm using methanol-water(77∶23)as the mobile phase.Results:Quercetin and piperine could be clearly detected by TLC.Quercetin and piperine had a good linear relationship between 6.25 and 200.The correlation coefficients of linear regression equation are 0.9995 and 0.9999 respectively.The average recovery rates of quercetin and piperine were 95.7%and 104.8%,respectively.Conclusion:This method is special,fast,simple and can be used to control the quality of Baras pill.
