摘要
目的观察糖肾清2号有效成分对糖尿病肾病小鼠肾脏及血清中PPARγ、RANTES、IL-1β、MCP-1水平的影响,探讨该方减轻DN小鼠肾脏损伤的作用。方法将KK-Ay小鼠30只予高脂饲料喂养3周制备糖尿病肾病模型。造模成功的小鼠随机分为模型组,阳性药组(缬沙坦13.33mg/kg),糖肾清2号高、中、低剂量组(糖肾清2号方有效成分13.33mg/kg,6.67mg/kg,3.33mg/kg),另设6只C57BL/6J小鼠为正常组,各组小鼠灌胃1次/d,连续干预12周后取血清及肾脏。采用RT-PCR技术检测肾脏组织PPARγ,RANTES,IL-1β,MCP-1mRNA转录水平,Western blot技术测定肾脏组织PPARγ、RANTES蛋白表达水平。ELISA法检测血清IL-1β、MCP-1蛋白表达水平。结果与正常组相比,模型组PPARγmRNA水平下调,蛋白表达水平上调(P<0.01),RANTES、IL-1β、MCP-1 mRNA水平及蛋白表达水平上调(P<0.01)。与模型组比较,TSQ高、中、低剂量组PPARγmRNA水平下调,蛋白水平上调(P<0.05,P<0.01),RANTES、IL-1β、MCP-1mRNA及蛋白表达均下调(P<0.05,P<0.01)。结论糖肾清2号方可以上调DN小鼠PPARγ水平,抑制炎性损伤,对DN有一定保护作用。
Objective To observe the effect of Tangshenqing(TSQ)2 active ingredient on the gene and protein expression level of PPARγ、RANTES、IL-1βand MCP-1 in kidney and serum of diabetic nephropathy mice and explore its molecular mechanism of inhibiting renal pathological damage.Methods Thirty KK-Ay mice were fed a high-fat diet for 3 weeks to prepare a diabetic nephropathy model.Mice successfully modeled with DN were randomly divided into model group,positive control group(Valsartan,13.33 mg/kg),TSQ high,medium and low dose groups(TSQ 2 active ingredient 13.33,6.67,3.33 mg/kg)with 6 mice in each group,another six C57BL/6J mice were used as normal group.Each group of mice was intragastrically administered once a day,and serum and kidney were taken after 12 weeks of continuous intervention.The mRNA levels of PPARγ,RANTES,IL-1βand MCP-1 were detected by RT-PCR.The expressions of PPARγand RANTES in renal tissues were determined by Western blot.The levels of serum IL-1βand MCP-1 protein were detected by ELISA.Results Compared with the normal group,the level of PPARγmRNA in model group was down-regulated,and protein level was up-regulated(P<0.01).RANTES,IL-1βand MCP-1 mRNA and protein expression levels were up-regulated(P<0.01).Compared with the model group,the PPARγmRNA levels in the high,medium and low dose groups of TSQ were down-regulated,the protein levels were up-regulated(P<0.05,P<0.01),and the expressions of RANTES,IL-1βand MCP-1 mRNA and protein levels were down-regulated(P<0.05,P<0.01).Conclusion Tangshenqing 2 can up-regulate the level of PPARγin DN mice,inhibit inflammatory damage,and have certain protective effect on DN.
