摘要
目的探讨组织生殖器形成抑制基因-1(suppressor with morphogenetic effect on genitalia-1,SMG-1)mRNA和SOX4mRNA检测在乳腺癌监测中的应用价值。方法回顾性分析2008年5月~2015年5月于咸阳市第一人民医院就诊的45例乳腺浸润性导管癌(IDC组)患者和32例乳腺非典型导管增生(ADH组)患者的临床资料,同时选择30例癌旁正常乳腺组织作为对照组,应用实时逆转录-聚合酶链反应(RT-PCR)技术检测组织SMG-1mRNA和SOX4mRNA的表达,比较分析两标志物的表达与乳腺癌的关系。结果组织SOX4mRNA在对照组、ADH组和IDC组中的异常率和表达水平分别为20.0%,53.1%,77.7%和0.21±0.07,0.57±0.13,0.89±0.18;组织SMG-1mRNA在三组中的异常率和表达水平分别为83.3%,60.0%,26.6%和0.92±0.21,0.62±0.12,0.25±0.09。与对照组比较,组织SOX4mRNA在ADH组与IDC组中的异常率和表达水平均显著增高,而SMG-1mRNA则显著降低;与ADH组比较,组织SOX4mRNA在IDC组中的异常率和表达水平显著增高,而SMG-1mRNA则显著降低;以上比较的差异均有统计学意义(F_(表达水平)=326.23~353.97,χ■=4.51~24.24,P<0.01或P<0.05)。血清CA153在对照组、ADH组和IDC组中的异常率和水平分别为3.3%,25.0%,44.4%和13.2±11.3,50.3±36.2,132.0±77.9μg/ml;血清CEA在三组中的异常率和表达水平分别为3.3%,12.5%,20.0%和2.3±1.6,19.3±7.6,47.3±31.9 ng/ml。与ADH组和对照组比较,血清CA153和CEA水平在IDC组中显著增高,其差异有统计学意义(χ~2=5.37~5.47,均P<0.01)。在45例乳腺癌患者中,组织SMG-1mRNA和SOX4mRNA表达具有负相关性(r=-0.832,P<0.01)。SMG-1mRNA表达与血清CA153和CEA水平有负相关性(r=-0.767,-0.623,P<0.01),而SOX4mRNA则有正相关性(r=0.779,0.605,P<0.01)。组织SOX4mRNA的表达与乳腺癌的肿瘤大小和淋巴结转移具有显著相关性(χ~2=5.37~5.47,均P<0.05)。SMG-1mRNA表达与乳腺癌的肿瘤大小、TNM分期、淋巴结转移及组织学分级均有相关性(均χ~2=4.60~6.03,P<0.05)。�
Objective To investigate the value of SMG-1mRNA and SOX4mRNA in monitoring breast cancer.Methods The clinical data of 45patients with invasive ductal carcinoma(IDC group)and 32patients with atypical ductal hyperplasia(ADH group)were retrospectively analyzed between May 2008and May 2015in the First People’s Hospital of Xianyang City,and 30cases of normal breast tissue adjacent to cancer were as control group.The expression of SMG-1mRNA and SOX4mRNA were detected by RT-PCR,and compared and analyzed the relationship with breast cancer.Results The positive rate and expression level of tissue SOX4gene in control group,ADH group and IDC group were 20.0%,53.1%,77.7%and 0.21±0.07,0.57±0.13,0.89±0.18,respectively.And those of tissue SMG-1gene in three groups were 83.3%,60.0%,26.6%and 0.92±0.21,0.62±0.12,0.25±0.09,respectively.Compared with those in the control group,the positive rate and expression of SOX4mRNA were significantly increased and which of SMG-1mRNA were significantly decreased in ADH and IDC groups.In addition,these similar presenses of SOX4mRNA and SMG-1mRNA had been described in the IDC group compared with in the ADH group(Fexpression=326.33~353.97,χabnormalrate=4.51~24.24,P<0.001or 0.05).The positive rate and the expression level of serum CA153in the control group,ADH group and IDC group were 3.3%,25.0%,44.4%and 13.2±11.3,50.3±36.2,132.0±77.9μg/ml,respectively,and those of serum CEA in the third group were 3.3%,12.5%,20.0%and 2.3±1.6,19.3±7.6,47.3±31.9ng/ml,respectively.The serumal levels of CA153 and CEA were observed to be significantly higher in the IDC group than those in the control and ADH groups(P<0.01).In 45cases of the patiens with breast cancer,the expression of SMG-1mRNA and SOX4mRNA was found to be negatively correlated(r=-0.832,P<0.01).Furthermore,here,showed these negative correlations between SMG-1mRNA expression and serum levels of CA153and CEA(r=-0.767,-0.623,P<0.01,respectively),and these positive correlations between SOX4mRNA expression and them(r=0.779,0.605,P<0.01,respe
