摘要
以牛樟带腋芽茎段为外植体,建立并优化了牛樟组培繁育体系,研究不同消毒处理、不同激素及配比对牛樟茎段不定芽诱导、丛芽增殖及瓶内生根的影响。结果表明:牛樟初诱导采用0.1%HgCl2+吐温-20消毒8 min污染率最低,腋芽正常萌发;牛樟带腋芽茎段最佳启动培养基配方为MS+6-BA 1.0 mg·L-1+IAA 0.5 mg·L-1,在此培养基中不定芽培养13 d开始萌动,诱导率可达80.00%;不定芽继代培养基的最适宜配方为MS+6-BA 1.0 mg·L-1,在此培养基中不定芽生长健壮,愈伤组织少,增殖系数达到了3.83,在继代培养基中添加0.2 g·L-1的活性碳有壮苗和防止褐化作用;幼苗生根培养基的最适宜配方为MS+6-BA 0.1 mg·L-1+IAA 0.5 mg·L-1+0.2 g·L-1 AC,在此培养基中幼苗培养18 d开始发根,根系发达,平均单株根数达4.23,生根率达82.50%。上述组织培养技术条件,可有效提高牛樟茎段的不定芽诱导率和生根率,为牛樟产业化育苗奠定了基础。
To establish a tissue culture system for Cinnamomum kanehirae,the stems with buds of3-year-old C.kanehirae tree were used as explants in vitro rapid propagation.Effects of different sterilization treatments and hormone on buds initiation,shoot proliferation and rooting of tissue cultured seedlings were analyzed.The results showed that the best sterilization method was0.1%HgCl2+Tween-20for8min.The optimal medium for inducing axillary buds was MS+1.0mg·L^-16-BA+0.5mg·L^-1IAA,in which the start time of adventitious bud induction was13d and the budding rate reached80.00%.The best medium for propagation of adventitious buds was MS+1.0mg·L^-16-BA,in which the adventitious buds grew vigorously with a proliferation rate of3.83.Adding0.2g·L^-1of AC(activated carbon)in the subculture medium helped to prevent browning and form strong seedlings.The best medium for rooting was MS+6-BA0.1mg·L^-1+IAA0.5mg·L^-1+0.2g·L^-1AC,in which start time of rooting was18d and the average root number of single plant was4.23with a rooting rate of82.50%.This optimized tissue culture condition effectively improves the introduction rate of adventitious buds and the rooting rate of aseptic seedlings,providing a technical foundation for industrialized seedlings of improved varieties of C.kanehirae.
