摘要
以24年生大叶栎优树环割萌芽条的嫩茎为外植体,用0.1%HgCl2消毒6 min,无菌活体得率40%;在BMT基本培养基上添加6-BA 0.3 mg/L和IBA 0.25 mg/L,芽继代培养25天,增殖系数3.9,且生长健壮;单芽在1/2 BMT+IBA 1.0 mg/L培养基上进行生根诱导培养,生根率80%,形成完整的组培再生植株。
24-year old superior trees are girdled to promote sprouting, and the tender stems are used as explants to study the tissue culture and plantlet regeneration of Castanopsis fissa. The results are as follows: The best effects are achieved when the explants are treated with 0.1% HgCl2 for 6 minutes, and the yield rate of aseptic in vitro is 40 %. The optimal subculture medium is BMT + 6-BA 0.3 mg/L + IBA 0.25 mg/L. The propagation coefficient could be up to 3.9/25d.The rooting medium for test-tube plantlets is 1/2BMT + IBA 1.0 mg/L, and the rooting rate reaches 80 %.
