摘要
无菌采集死亡羔羊肺脏样品接种于含100mL/L马血清的TSA平板,37℃培养24h^48h分离病原菌,观察分离菌菌落特征、染色特性,分析其溶血性等,采用山羊肺炎常见病原菌的特异性引物,以无菌水稀释菌落,微波炉热处理稀释菌落释放的细菌基因组DNA为模板,PCR扩增病原菌目的基因片段鉴定分离的病原菌,通过药敏试验筛选敏感药物。结果表明,分离鉴定出2种病原菌分别为多杀性巴氏杆菌和溶血性曼氏杆菌,其中溶血性曼氏杆菌呈β溶血;分离鉴定的2种病原菌均能分别致死Balb/c小鼠;2株分离菌对头孢噻肟、头孢唑林、头孢拉定、头孢美唑、庆大霉素、丁胺卡那、诺氟沙星、左氧氟沙星、氧氟沙星、四环素和多西环素共11种抗菌药物高度敏感。表明多杀性巴氏杆菌和溶血性曼氏杆菌是致该病例山羊羔死亡的重要病原菌,为该羊场山羊细菌性肺炎的防控提供了依据。
The aim of this study was to find pathogenic bacteria of fatal pneumonia in a goat.This experiment isolated and identified pathogenic bacteria from the lungs of a dead goat lamb and analyzed their haemolysis,pathogenicity and sensitivity to antibacterial drugs.The lung sample of one dead goat lamb was aseptically collected and inoculated on TSA plates containing 100 mL/L horse serum,and cultured at 37℃for 24 hours to 48 hours.Then,the colony characteristics and dyeing characteristics of isolated bacteria were observed.In order to further identify the isolated pathogenic bacteria,the bacteria,diluted by aseptic water,were heated in a microwave oven to release DNA.The target genes were amplified by specific primer PCR of Pasteurella multocida and Mannheimia haemolytica.The results showed that two strains of isolated bacteria were identified,which were Pasteurella multocida andβhaemolyisis Mannheimia haemolytica.Drug susceptibility test results showed that the isolated two strains were highly sensitive to cefotaxime,cefazolin,cefradine,cefmetazole,gentamicin,amikacin,norfloxacin,levofloxacin,ofloxacin,tetracycline and doxycycline.This study clarified some biological characteristics of the two bacterial pathogens and provided the basis for the prevention and control of bacterial pneumonia in the goat farm.
