摘要
目的建立测定解郁安神颗粒中柴胡皂苷含量的高效液相色谱(HPLC)法。方法色谱柱为Thermo Hypersil BDS C18柱(250 mm×4.6 mm,5μm),流动相为水-乙腈,梯度洗脱,检测波长为210 nm,流速为1.0 m L/min,柱温为25℃。结果柴胡皂苷a的质量浓度线性范围为20.25~311.46μg/m L,r=0.999 7(n=7),平均回收率为95.31%,RSD为1.29%(n=6);柴胡皂苷d的质量浓度线性范围为23.12~308.56μg/m L,r=0.999 6(n=7);平均回收率为96.35%,RSD为1.38%(n=6)。结论该法能同时测定解郁安神颗粒中柴胡皂苷a和柴胡皂苷d的含量,结果准确、重复性好、可操作性强,为加强以柴胡皂苷作为有效成分的中成药质量控制提供了参考。
Objective To establish an HPLC method for content determination of saikosaponin in Jieyu′anshen Granules.Methods The column was Thermo Hypersil BDS C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-water with gradient elu-tion,the detection wavelength was 210 nm,the flow rate was 1 mL/min,and the column temperature was 25℃.Results The linearity of saikosaponin a within the range of 20.25-311.46μg/mL(r=0.999 6),the average recovery was 95.31%,the RSD was 1.29%.The linearity of saikosaponin d within the range of 23.12-308.56μg/mL(r=0.999 7),the average recovery was 96.35%,the RSD was 1.38%(n=6).Conclusion The method can simultaneously determine the content of saikosaponin a and saikosaponin d in Jieyu′anshen Granules.The method is accurate,reproducible and operable,which can provide reference for quality control of saikosaponin as an effective component of Chinese patent medicine.
