摘要
目的建立柴桂解郁颗粒的质量标准。方法薄层色谱法对方中的柴胡、黄芩、干姜、大黄进行定性鉴别;高效液相法测定方中柴胡皂苷a的含量,色谱条件:色谱柱:Agilent ZORBAX SB-C18(4.6 mm×250 mm,5-Micron),流动相为乙腈-水(34:66),检测波长205 nm,柱温35℃。结果建立的薄层色谱中斑点清晰,阴性无干扰;柴胡皂苷a在浓度0.041~0.656 μg范围内线性良好(r=0.9999),平均回收率为100.29%,RSD为0.43%。结论本方法简便可行,重现性好,针对性强,可以很好地控制柴桂解郁颗粒的质量。
Objective To establish the quality standard of Chaigui Jieyu Particles.Methods The thin layer chromatography(TLC)method was used for qualitative identification of Chaihu(Chinese thorowax root),Huangqin(baical skullcap root),Ganjiang(zingiber)and Dahuang(rhubarb root and rhizome)in the prescription.High performance liquid phase(HPLC)was used to determine the content of saikosaponin a.Chromatographic conditions:Agilent ZORBAX SB-C18(4.6 mm×250 mm,5-Micron),mobile phase acetonitrile-water(34:66),detection wavelength 205 nm,column temperature 35°C.Results The establishment of TLC spots clear,negative with no interference;the linearity of saikosaponin a was good in the concentration range of 0.041-0.656μg(r=0.9999);the average recovery rate was 100.29%,and RSD was 0.43%.Conclusion This method is simple and feasible,reproducible,and highly targeted,and can well control the quality of Chaiui Jieyu Particles.
