解郁开心丹对抑郁模型大鼠神经内分泌的影响 被引量:8

摘要 目的观察解郁开心丹对抑郁模型大鼠血清皮质醇、β-内啡肽含量的影响。方法将80只SD雄性大鼠随机分为正常组、模型组、阳性药物对照组、中药高剂量组、中药中剂量组、中药低剂量组,每组10只;用慢性应激结合孤养的方式造模。造模后给药,阳性药物对照组给予盐酸氟西汀混悬液10 mg/d;中药高、中、低剂量组分别给予解郁开心丹2.0,1.0,0.5 g/d(给药体积为每100 g体质量给予1.0 m L),模型组给予等量生理盐水,连续28 d。用放射免疫分析法检测大鼠血清皮质醇、β-内啡肽的含量。结果与模型组比较,中药高剂量组、中药中剂量组大鼠的血清皮质醇含量明显减少,β-内啡肽的含量明显增加,P<0.05;中药高剂量组、中药中剂量组与阳性药物对照组比较无明显差异,P>0.05。结论解郁开心丹具有与西药抗抑郁药物氟西汀同样的抗抑郁作用,且抗抑郁作用机制可能与调节大鼠的血清皮质醇、β-内啡肽的平衡有关。 Objective To observe the influence of Jie Yu Kai Xin Dan on depression model rats' serum cortisol and the content of beta-endorphin. Methods The 80 male SD rats were randomly divided into normal group,model group,positive drug control group,Chinese medicine high dose group,Chinese medicine medium dose group,Chinese medicine low dose group,each group of 10 rats. Make mould with chronic stress combining with solitary keeps the way. After building,positive drug control group given fluoxetine hydrochloride mixed suspension 10 mg / d. Chinese medicine high,medium and low dose groups,respectively,to give Jie Yu Kai Xin Dan 2. 0 g,1. 0 g and 0. 5 g / d( dosing volume per 100 g body quality give 1. 0 m L),model group was given amount of normal saline,continuous 28 d. Radiation immunity analysis method is used to inspect the rat serum cortisol,the content of beta-endorphin. Results Compared with model group,high dose of Chinese Medicine group and medium dose group of rats serum cortisol content decreased significantly,the content of beta-endorphin increased significantly,P〈0. 05. Traditional Chinese medicine( TCM) in high dose group and medium dose group compared with positive drug control group has no significant difference,P〈0. 05. Conclusion Jie Yu Kai Xin Dan has the same antidepressant effect with western medicine antidepressants fluoxetine,and the antidepressant mechanism may be related to regulating the rat serum cortisol and beta-endorphin of balance.
