骨髓间充质干细胞移植修复皮肤创伤 被引量:11

摘要 背景:目前大部分有关间充质干细胞治疗皮肤创伤的实验均使用大鼠、小鼠和兔进行,而应用犬骨髓间充质干细胞治疗皮肤创伤的研究较少。目的:观察犬骨髓间充质干细胞治疗皮肤创面的效果。方法:在4只犬肩胛部、臀部两侧分别做3 cm×3 cm的正方形皮肤创伤,右侧作为实验组,在创面制作后的第1,3天,创口皮下注射异体犬骨髓间充质干细胞悬液;左侧作为对照组,在创面制作后的第1,3天,创口皮下注射间充质干细胞培养基,动态观察创面愈合情况。结果与结论:在第1周时间内,两组创口处都有淡黄色炎性物质存在,炎症反应明显,且对照组炎性物质较实验组多,肉芽组织生长速度慢于实验组;第2周至伤口愈合时间段,创口在形成肉芽组织的基础上开始明显上皮化,主要表现从伤口四周向中央形成新鲜上皮,实验组开始上皮化时间早于对照组,创口面积小于对照组;第3周时,实验组有的创口完全愈合,且愈合后的创面较对照组平整、光滑。实验组细胞移植后8,12 d的创口面积略小于对照组,创口愈合速度稍快于对照组,但差异均无统计学意义。结果表明,犬骨髓间充质干细胞移植具有促进皮肤创面愈合的可能或趋势。 BACKGROUND:At present,most of the experiments on the treatment of skin wound by mesenchymal stem cellshave been performed in rats,mice and rabbits,while the research on skin wound treatment by canine bonemarrow mesenchymal stem cells is less reported.OBJECTIVE:To observe the effect of canine bone marrow mesenchymal stem cells on skin wound healing.METHODS:A3cm×3cm wound was made on the both sides of the scapula and buttocks of the dog,with the rightside as experimental group and the left side as control group.After the wound was made,allogeneic canine bonemarrow mesenchymal stem cells suspension was injected subcutaneously around the wound in the experimentalgroup on the1st and3rd days.The control group was injected subcutaneously around the wound withmesenchymal stem cell culture medium on the1st and3rd days after the wound was made.Wound healing wasobserved dynamically in both groups.RESULTS AND CONCLUSION:In the1st week,there were pale yellow inflammatory substances in the wound oftwo groups indicating obvious inflammations.Compared with the control group,the inflammatory substances were fewer and the growth rate of the granulation tissue was faster in the experimental group.From the2nd week until thewound healing,epithelialization on the wound became obvious following the formation of the granulation tissue,whichwas mainly displayed by the formation of fresh epithelial tissues from the surrounding to the wound center.Theepithelialization time of the experimental group was earlier than that of the control group,and the wound area of theexperimental group was smaller than that of the control group.In the3rd week,the wound in the experimental grouphealed completely,and became smoother than that in the control group.The wound area of the experimental group wasslightly smaller than that of the control group on the8th and12th days after cell transplantation,and the healing speed ofthe experimental group was slightly faster than that of the control group,but there was no significant difference betweenthe two groups
