摘要
目的:研究NF-κB基因沉默对人胃癌细胞SGC7901增殖及TFF3、ERK表达的影响。方法:应用脂质体法将靶向NF-κB的干扰RNA(si RNA)转染入胃癌细胞株SGC7901中。采用MTT法和流式细胞术法检测NF-κB基因沉默对SGC7901细胞的增殖抑制率和细胞凋亡情况,运用Real time PCR和Western blot免疫蛋白印迹法检测SGC7901细胞内TFF3、ERK的表达。结果:MTT法显示NF-κB基因沉默组癌细胞抑制率大于空白载体组和对照组(P〈0.01);流式细胞术法表明NF-κB基因沉默组细胞凋亡比例达12.60%,明显高于空白载体组和对照组(P〈0.01);RT-PCR和Western Blot检测示,经NF-κB基因沉默后,细胞内NF-κB、TFF3、ERK m RNA及蛋白表达量下降明显。结论:NF-κB基因沉默可抑制胃癌细胞的增殖并促进其凋亡。NF-κB基因作为胃癌治疗的分子靶点可能具有重要意义。
Objective:To study the effects of NF-κB gene silencing on the proliferation and expression of TFF3 and ERK in human gastric cancer cell line SGC7901. Methods:RNA(si RNA)was transfected into gastric cancer cell line SGC7901 by liposome targeting NF-κB. MTT method and flow cytometry were used to detect the proliferation inhibition and apoptosis of NF-κB gene silencing in SGC7901 cells,the m RNA and protein expression level of TFF3,ERK were detected by Real time PCR and Western blot. Results:MTT showed that the inhibition rate of NF-κB gene silencing group was higher than that of the blank control group and the control group(P〈0.01);Flow cytometry showed that the percentage of apoptosis in the NF-κB gene silencing group was 12.60%,higher than that in the blank control group and the control group(P〈0.01);RT-PCR and Western Blot detection,The NF-κB,TFF3,ERK m RNA and protein expression in the cells decreased significantly after NF-κB gene silencing. Conclusion:NF-κB gene silencing can inhibit the proliferation of gastric cancer cells and promote its apoptosis,NF-κB gene may be of great significance as a molecular target for the treatment of gastric cancer.
