摘要 基于茶多酚在碱性环境中对Luminol-H_2O_2-AuNPs化学发光体系有明显的抑制现象,建立一种快速检测茶多酚含量的化学发光新方法。采用单因素法和响应曲面优化对化学发光体系中的Luminol、H_2O_2、NaOH、纳米金浓度进行优化,用Design-Expert中的Trial对所得的结果进行二次回归拟合并得到数学模型,对该模型的相关各参数进行方差分析及统计,结果表明该结论可以用于茶多酚含量的预测。在最优实验条件下茶多酚的浓度和发光强度在1.0×10^(-7)~1.0×10^(-5) g/L范围内具有良好的线性关系,其检出限为8.0×10^(-8) g/L,相对标准偏差为2.4%,此方法可用于茶叶中茶多酚含量的检测,并对其可能的化学发光机理进行了探讨。 Because tea polyphenol can strongly restrict Luminol-H2 O2-AuNPs chemiluminescence in alkaline medium, a highly sensitive chemiluminescence(CL) method for the determination of tea polyphenol was established based on the phenomenon. The effect of luminol concentration, H2 O2 concentration, NaOH concentration, AuNPs concentration on the luminescence of the system was investigated by single factor optimization and response surface optimization methods. The quadratic regression fitting of the result was carried out by the Trial of the Design-Expert, and the mathematical model was obtained, and analysis of variance and statistics of related parameters of the model were carried out by least square method. The experimental results showed that the conclusion can be used to predict tea polyphenols content. Under the optimal experimental conditions, the relative CL intensity and the tea polyphenols concentration showed a good linear relationship in the range of 1.0×10^(-7)~1.0×10^(-5) g/L, the detection limit was 8.0×10^(-8) g/L, relative standard deviation was 2.4%.This method can be used in the detection of tea polyphenol content in tea. At last, the possible chemical luminescence mechanism was discussed.
