摘要
目的研究木兰脂素对肺癌的影响,并探讨其对顺铂的增敏作用。方法用Annexin V/PI凋亡检测试剂盒检测木兰脂素体外增敏顺铂诱导肺癌细胞凋亡等能力;用平板克隆形成实验验证木兰脂素及顺铂对肺癌细胞克隆形成能力的影响。将40只BALB/c裸鼠分为空白组(生理盐水)、对照组Ⅰ(单用木兰脂素)、对照组Ⅱ(单用顺铂)及实验组(顺铂及木兰脂素联用)。观察小鼠的一般情况,记录肿瘤重量,计算肿瘤抑制率及增效率;通过免疫组化法检测小鼠肿瘤Ki-67及血管内皮生长因子(VEGF)的表达情况。结果空白组、对照组Ⅰ、对照组Ⅱ及实验组的总细胞凋亡率分别为(7.53±1.25)%,(15.84±1.45)%,(29.47±2.15)%,(76.24±4.12)%,克隆形成的抑制率分别为(5.45±0.87)%,(18.55±2.34)%,(28.54±3.25)%,(93.54±5.64)%,差异均有统计学意义(均P<0.05)。空白组及对照组Ⅰ小鼠的生存质量最好,其次是实验组,对照组Ⅱ小鼠的生存质量最差,药物不良反应最明显。空白组、对照组Ⅰ、对照组Ⅱ及实验组的抑瘤率分别为(1.54±0.67)%,(8.56±1.23)%,(41.89±2.12)%,(54.38±0.78)%,肿瘤Ki-67的表达率分别为(67.43±2.12)%,(52.38±3.21)%,(37.67±0.45)%,(23.54±0.34)%,VEGF的表达率分别为(21.34±2.72)%,(17.34±3.12)%,(14.34±2.45)%,(9.43±1.23)%,差异均有统计学意义(均P<0.05)。结论木兰脂素能够促进顺铂的抗肿瘤作用,其作用机制可能与抑制VEGF的表达有关。
Objective To study the effect of Magnolin in the treatment of lung cancer mice and to explore its sensitizing effect on cisplatin.Methods The apoptosis of lung cancer cells induced by cisplatin in vitro was detected by Annexin V/PI apoptosis detection kit. The effect of magnolacin and cisplatin on the colony formation of lung cancer cells was verified by plate colony formation assay. Forty BALB/c nude mice were divided into blank group( normal saline),control group Ⅰ( Magnolin only), control group Ⅱ( cisplatin alone) and experimental group( cisplatin combined with Magnolin). To observe the general condition of the mice,record the tumor weight,calculate the tumor inhibition rate and efficiency,and to detect the expression of Ki-67 and vascular endothelial growth factor( VEGF) in mice by immunohistochemistry. Results The apoptotic necrosis rates in blank group,control group Ⅰ,control group Ⅱ and experimental group were( 7. 53 ± 1. 25) %,( 15. 84 ± 1. 45) %,( 29. 47 ± 2. 15) %,( 76. 24 ± 4. 12) %. The inhibition rates of colony formation were( 5. 45 ± 0. 87) %,( 18. 55 ± 2. 34) %,( 28. 54 ± 3. 25) % and( 93. 54 ± 5. 64) %,all with significant difference. The quality of life of the mice in blank group and control group Ⅰ was the best,followed by experimental group. The mice in control group Ⅱ had the worst quality of life and the adverse drug reactions were the most obvious.
