亚硒酸钠诱导肺癌A549细胞凋亡的作用及对凋亡相关蛋白Cytc影响

摘要 目的探讨凋亡相关蛋白Cytc是否与亚硒酸钠诱导肺癌细胞凋亡有关。方法 A549人肺癌细胞株按照亚硒酸钠浓度(0μmol/L,0.5μmol/L,2.5μmol/L,5.0μmol/L,8.0μmol/L)由低到高分为5组,加相应浓度的亚硒酸钠培养,噻唑蓝(MTT)法检测亚硒酸钠对A549细胞的抑制率;凋亡相关蛋白Cytc在A549细胞分布通过免疫细胞化学染色法检测,免疫印迹法检测亚硒酸钠对Cytc蛋白含量的影响,TUNEL检查亚硒酸钠对肺癌A549凋亡的影响。结果 MTT法结果显示:人肺癌A549细胞株,随着亚硒酸钠浓度的增加,其吸光度值逐渐降低,仅呈现浓度依懒性(P<0.01);亚硒酸钠对肺癌A549细胞株的生长抑制率2.5μmol/L和5μmol/L亚硒酸钠组呈现浓度和时间依赖性(P<0.05),0.5μmol/L和8μmol/L亚硒酸钠组仅呈现浓度依赖性(P<0.05)。免疫组织化学和Western blot印迹结果均显示Cytc蛋白在肺癌A549细胞中的表达随着亚硒酸钠浓度的增高逐渐升高(P<0.05)。TUNEL染色显示:亚硒酸钠诱导肺癌A549凋亡呈现浓度依赖性(P<0.05)。结论随着亚硒酸钠浓度升高肺癌A549细胞的抑制率升高,凋亡相关蛋白Cytc的表达也升高,故亚硒酸钠浓度诱导肺癌A549细胞凋亡与Cytc有关。 Objective: To investigate the role of sodium selenite in inducing apoptosis of lung cancer cells and its molecular mechanism. Methods: Human lung cancer A549 cell lines were divided into blank control group,0μmol/L0. 5μmol/L,2. 5μmol/L,5. 0μmol/L and 8. 0 μmol/L sodium selenite group. After incubation with different concentrations of sodium selenite for 24 h and 48 h,MTT assay was used to detect sodium selenite. The effect of sodium selenite on the expression of Cytc was examined by immunocytochemical staining and Western blotting. The effect of sodium selenite on the apoptosis of lung cancer A549 was examined by TUNEL assay. Results: The results of MTT assay showed that human lung cancer cell line A549 was cultured with different concentrations of sodium selenite for 24 h and 48 h. The absorbance value gradually decreased with the increase of sodium selenite concentration,showing only concentration dependence( P〈 0. 01); sodium selenite inhibited the proliferation of lung cancer A549 cell lines at a concentration of 2. 5 μmol/L and 5μmol/L sodium selenite group in a concentration-time dependent manner( P〈 0. 05),0. 5 μmol/L and 8 μmol/L sodium selenite group showed only concentration dependent( P〈 0. 05). The results of immunohistochemistry and Western blot showed that the expression of Cytc protein in lung cancer A549 cells increased gradually with the increase of sodium selenite concentration( P〈 0. 05). TUNEL staining showed that sodium selenite induced apoptosis of lung cancer A549 in a concentration-dependent manner( P〈 0. 05). Conclusion: These results suggest that sodium selenite can inhibit the growth of lung cancer A549 cells and induce apoptosis,which may be related to the release of mitochondrial Cytc.
